| Polysaccharides are important active ingredients of Sendeng-4 and its single herb medicines.Many studies have shown that the polysaccharide can enhance immune function,lowering blood pressure,lowering blood sugar,anti-tumor,anti-bacterial,anti-viral,anti-radiation,anti-clotting,anti-aging,anti-oxidation.Mongolian medicine Sendeng-4 and single herbs are commonly used drugs,which are used for the treatment of anti-rheumatic and other diseases.According to the literature,at present,researches on polysaccharides from Sendeng-4 and its single herbs have not been comprehensive enough.Therefore,studies on extraction technology and antioxidant effect of polysaccharides from Sendeng-4 and its single herbs would be of great significance for the secondary development of Sendeng-4.In this paper,as polysaccharide content an indicator,solid-liquid ratio,extraction time,extraction temperature and extraction times were used for factors,optimization of extraction process of polysaccharides from Sendeng-4 and its single herbs in prescription by response surface design.In addition,the scavenging activities of the polysaccharides from Sendeng-4 and its single herbs in prescription on hydroxyl,superoxide radicals,nitrite,DPPH and the reducing capacity were investigated,and then compared oxidation activities of Sendeng-4before and after in compatibility.The results of study on the extraction process of polysaccharides showed that the optimum extraction conditions of Sendeng-4 and single herbs were different.(1)The optimum extraction conditions of polysaccharides from Sendeng-4 were : solid-liquid ratio1:40,extraction temperature of 95 ℃,the extraction time was 3 h,extracted for 3 times.At this conditions,extraction rate of polysaccharides reached 8.90%.(2)The optimum extraction conditions of polysaccharides from Wen-guan-mu were as follows: solid-liquid ratio 1:32,extraction temperature 69 ℃,extraction time of 1.8 h,extraction times 2 times,the yield of polysaccharides reached 1.45%.(3)The optimum extraction of Toosendan polysaccharides were: solid-liquid ratio 1:20,extraction temperature 90 ℃,extraction time of 2.9 h,extracted for 3 times,polysaccharides yield was 22.34%.(4)The optimum extraction process of Gardenia polysaccharide was: liquid ratio of 1: 33.7,the extraction temperature is 80 ℃,extraction time of 3 h,extracted for 3 times,the polysaccharides yield was 10.97%.(5)The optimum extraction chebulinic polysaccharide were:solid-liquid ratio 1:28,extraction temperature 69 ℃,extraction time was 1 h,extracted for3 times,the polysaccharides yield was 5.99%.Studies on antioxidation of polysaccharides showed that the antioxidant activitiesof polysaccharides from herbs have changed after the compatibility,and antioxidant activities of polysaccharides from Sendeng-4 is better than that of its four single herb.(1)The scavenging ·OH-activity of polysaccharides from Sendeng-4 was stronger than that of the reference Vc,Wenguanmu,Zhizi,and Chuanlianzi,weaker than that of the Hezi.Where the scavenge ability of polysaccharides from Zhizi up to 26.35%.(2)The scavenging O2-ability of polysaccharides from Sendeng-4 was stronger than that of the reference mannitol and other four herbs.(3)The scavenging NO2-ability of polysaccharides from Sendeng-4 was stronger than that of the reference Vc,Hezi,Chuanlianzi,weaker than that of the Zhizi,and this ability was same as Wenguanmu.(4)The scavenging DPPH ability of polysaccharides from Chuanlianzi and Hezi weaker than that of Vc,scavenging DPPH activity of polysaccharides from Wenguanmu and Sendeng-4were failed measured,respectively.(5)The reducing abilities of polysaccharides from Sendeng-4 and other four herbs were weaker than that of the reference Vc,and one of polysaccharides from Sendeng-4 also weaker than that of other four herbs. |
