Objective: To study the allele frequencies and the genotype distribution of transcription factor FOXP3 gene rs3761548 single nucleotide polymorphisms in normal populations,prediabetes,type Ⅱ diabetes,dyslipidemia and type Ⅱ diabetes combined dyslipidemia.To analyze the relationship between transcription factor FOXP3 gene rs3761548 single nucleotide polymorphisms and the risk of normal populations,prediabetes,type Ⅱdiabetes,dyslipidemia and typeⅡ diabetes combined dyslipidemia.Methods:(1)The Objects studied were all the Han people of Luzhou city in Si Chuan Province without blood relationship between objects and more than 40 old years.The all cases studied are 1300 objects,which is divided the control group(216 cases)and the disease group(1084 cases).The mono-factorial groups in the disease group consist of the prediabetes group(207cases),the type Ⅱ diabetes group(216 cases)and the dyslipidemia group(216 cases).The two-factor groups consist of the prediabetes combined dyslipidemia group(216 cases)and the type Ⅱ diabetes combined dyslipidemia group(229 cases).The 216 cases in the control group are without history symptoms and signs of dyslipidemia,diabetes and Hypertension.(2)The genomic DNA were extracted from the blood of all cases studied.The FOXP3 gene rs3761548 fragments were amplificated by polymerase chain reaction(PCR)and the PCR fragments were sequenced and digested by endonucleases Pst I.The genotypes were analyzed after the electrophoresis of the digested products.(3)The analysis of statistics(SPSS22.0 software): The genotype in population was detected by the Hardy-Weinberg law of equilibrium.The allele frequencies and the genotype distribution of transcription factor FOXP3 gene rs3761548 single nucleotide polymorphisms in different groups were analyzed by fit chi-square.Results:(1)The genotype and allele frequency showed that the FOXP3 gene rs3761548 single nucleotide polymorphism in the population was consistent with Hardy-Weinberg law of equilibrium(P>0.05).The genotype and allele frequency of Chinese Han people in Luzhou city is similar as that of other Chinese,Japanese and Mexicans(the date is from the NCBI databank).The genotype CC and the C allele frequency of the Chinese Han people in Luzhou city were decreased compared with the Europeans and the genotype CC and the C allele frequency of Chinese Han people in Luzhou city were increased compared with the Europeans.The genotype AA and the A allele frequency of the Chinese Han people in Luzhou city were increased compared with the Africans and the genotype CC and the C allele frequency of Chinese Han people in Luzhou city were decreased compared with the Africans.(2)There were no statistically significant for the genotype and allele frequency of FOXP3 gene rs3761548 in the control group compared with prediabetes group,dyslipidemia group and the prediabetes combined dyslipidemia group(P>0.05).(3)There was statistically significant for the genotype distribution of FOXP3 gene rs3761548 between the control group and type 2 diabetes.(χ2=11.909,P=0.003).(4)There was statistically significant about the genotype distribution of FOXP3 gene rs3761548 between the control group and typeⅡdiabetes combined dyslipidemia group(χ2=11.663,P=0.003).(5)There was no statistically significant for the genotype and allele frequency of FOXP3 gene rs3761548 between the type Ⅱ diabetes and type Ⅱ diabetes combined dyslipidemia group.(P>0.05)(6)There was statistically significant for the genotype and allele frequency of FOXP3 gene rs3761548 between the dyslipidemia group and type Ⅱ diabetes combined dyslipidemia group.(χ2=6.604,P=0.010,χ2=26.977,P=0.000)Conclusion:(1)The genotype and allele frequency of FOXP3 gene rs3761548 fragment in Chinese Han people of Luzhou city in Sichuan province is similar as other Chinese Han people,Japanese and Mexicans,but is different from that of Europeans and Africans.(2)There are no relationships between the FOXP3 gene rs3761548 gene single nucleotide polymorphism in control group and the prediabetes group,dyslipidemia group and prediabetes combined dyslipidemia group.(3)The FOXP3 gene rs3761548 gene single nucleotide polymorphism is interrelate with typeⅡiabetes.And the genotype AA an AC are the risk factors for type Ⅱ diabetes.(4)The FOXP3 gene rs3761548 gene single nucleotide polymorphism is interrelate with type Ⅱ diabetes combined dyslipidemia group.And the genotype AA an AC are the risk factors for type Ⅱ diabetes combined dyslipidemia group.
