Objective To investigate Myocyte-specific enhancer binding factor 2A (MEF2A) expression in temporal neocortex tissue samples from patients with intractable epilepsy and in hippocampus of the lithium-pilocarpine-induced TLE rat model.Methods1.20 temporal neocortex tissue from patients with intractable epilepsy and 20 histological normal neocortex tissue were selected.2. Adult male Sprague-Dawley rats were divided randomly into seven groups, including six groups with epilepsy induced by lithium-pilocarpine administration and one control group without epilepsy. And then hippocampal tissues were taken from rats at 1d,3d, 1w,2w, 1m,2m after seizures, and from controls.3.Expression of MEF2A were dectected by double-label immunofluorescent staining, immunohistochemistry, real-time quantitative PCR and western blot.Results1. MEF2A was found to be expressed in the nuclei of neurons but not in the dendrites of neurons and astrocytes by double immunofluorescence labeling.2. By the use of immunohistochemistry staining, the faint and scattered immunoreactivity for MEF2A was observed in the nuclei of neuron in the temporal neocortex of patients with TLE and the in hippocampus of TLE model,whereas strong immunoreactivity in the controls.3. The qRT-PCR results showed that MEF2A mRNA level was found to be decreased in TLE patients, as compared to controls.And in the lithium-pilocarpine-induced TLE model, MEF2A expression dynamically decreased within 2 months. The lowest level of MEF2A expression occurred at day 7.4. The western blotting result was in accordance with qPCR. Strongly stained bands were present in all control group samples, while relatively faint expression of the MEF2A was observed in the TLE group. In addition,MEF2A expression in the hippocampus of TLE rats was transiently decreased at 1day after seizure and The lowest level of MEF2A expression occurred at 1w.Conclusions MEF2A may involve in the pathogenesis of TLE in regulation of synaptic plasticity and neuronal apoptosis.
