Objective: we study the expression and correlation of activation of pancreatic stellate cells(PSC) marker alpha smooth muscle actin(alpha-smooth muscle actin,alpha SMA) and Hic-5 in pancreatic cancer and its relationship with the clinical pathological characteristics of pancreatic cancer,to investigate the role of activated pancreatic stellate cells with each other, providing a new target for the treatment of pancreatic cancer and theoretical basis.Methods:1.Isolation and culture of normal mouse PSC,immunofluorescence staining was used to observe PSC,using Western blot to detect the expression levels of Hic-5 and alpha–SMA;2.Taking human pancreatic cancer tissue and normal pancreatic tissue were identified by immunohistochemical staining with alpha-SMA and Hic-5;3. Clinical follow up of all patients,TNM stage and survival time and other factors of the patients were statistically calculated,analyze the relationship between the expression of alpha-SMA and Hic-5 in pancreatic carcinoma and their relationship with the clinical pathology.Results: 1.The expression of alpha-SMA and Hic-5 in human pancreatic cancer tissue was significantly higher than that in the normal pancreas group;2.In normal mouse pancreatic tissue,alpha-SMA and Hic-5 are expressed in activated pancreatic stellate cells,and it was significantly higher than that of the pancreatic stellate cells in the quiescent stage;3. The expression degree of activated pancreatic stellate cells and Hic-5 were correlated with the classification,tumor size,prognosis of pancreatic cancer. Conclusion: 1.Isolate and culture of primary mouse pancreatic stellate cells successfully;2. The expression of Hic-5 in the active phase / pancreatic cancer was significantly higher than in the quiescent stage / normal pancreatic tissue,indicating that pancreatic cancer tissue promotes the activation of pancreatic stellate cells;3. Hic-5 and pancreatic stellate cells have an important role of tumor size,differentiation degree and so on in human pancreatic cancer,and may be related to the survival time.
