Objective To investigate the effect of apolipoprotein E mimetic peptide COG 1410 on the early cerebral edema,Aquaproins-4(AQP-4)expression and movement function in mice in the early brain injury(EBI)after subarachnoid hemorrhage(SAH).Methods SAH model was induced by endovascular perforation.A total of 108 Male C57BL/6J mice were randomLy divided 36 mice into sham injury group,36 mice into SAH+saline group and 36 mice into SAH+COG1410 group.We are randomly selected 6 mice from control group,6 mice from saline group and 6 mice from sham group which are used to observe the mice motion function change after SAH.We are randomly selected 6 mice from control group,6 mice from saline group and 6 mice from sham group which are used to observe the mice brain water content after SAH.We are randomly selected 6 mice from control group,6 mice from saline group and 6 mice from sham group which are used to observe the mice the level of AQP-4 protein after SAH.We are randomly selected 6 mice from control group,6 mice from saline group and 6 mice from sham group which are used to observe the mice the number of positive cells of AQP-4 after SAH.We are randomly selected 6 mice from control group,6 mice from saline group and 6 mice from sham group which are used to observe the mice AQP-4mRNA expression after SAH.Results After SAH nerve function score saline group and treatment group were significantly higher than the control group(P<0.05),and there are differences between normal saline group and treatment group(P<0.05),The nerve function score increased in 1d after SAH and reached the peak at 2 d,then gradually decline.After SAH brain water content saline group and treatment group were significantly higher than the control group(P<0.05),and there are differences between normal saline group and treatment group(P<0.05),The brain water content increased in 1d after SAH and reached the peak at 2 d,then gradually decline.After SAH the level of AQP-4 protein saline group and treatment group were significantly higher than the control group(P<0.05),and there are differences between normal saline group and treatment group(P<0.05),The level of AQP-4 protein increased in 1d after SAH and reached the peak at 2 d,then gradually decline.After SAH the number of positive cells of AQP-4 saline group and treatment group were significantly higher than the control group(P<0.05),and there are differences between normal saline group and treatment group(P<0.05),The number of positive cells of AQP-4increased in 1d after SAH and reached the peak at 2 d,then gradually decline.After SAH AQP-4mRNA expression saline group and treatment group were significantly higher than the control group(P<0.05),and there are differences between normal saline group and treatment group(P<0.05),AQP-4mRNA expression increased in 1d after SAH and reached the peak at 2 d,then gradually decline.Conclusion Administration of COG1410 significantly inhibited cerebral edema via inhibition of AQP-4 expression.
