Objective:To observe the expressions of retinal angiogenesis regulation factors of diabetes mice and investigate the effects of the method of activing blood,Tonifying qi and nourishing yin expressions of retinal angiogenesis regulation factors of diabetes mice. Thereby researching the protective mechanism of diabetic retinopathy.Methods:Diabetes mellitus (DM) was induced in 90 male C57BL/6 mice (C57 mice) by peritoneal injection of streptozotocin. The 90 mice were randomly selected from 101 male C57 mice. After 2 months,58 diabetes mellitus model mice(fasting blood glucose>13.9 mmol/L) were randomly divided into high-dosage group, middle-dosage group, low-dosage group, positive drug control group and DM model group by using SPSS. The experimental drug was Compound Xueshuantong Capsule (XST).The positive drug control drug was calcium dobesilate. After a 8-week intervention, retinal tissues were taken in every groups and the expressions of retinal vascular endothelial growth factor (VEGF)、pigment epithelium derived factor (PEDF)、insulin-like growth factor-1 (IGF-1) were observed using immunohistochemical staining. The results were analyzed by SPSS 17.0.Results:1. Compared with normal control group, the statistics result of the expression of VEGF were higher in DM model group and low-dosage group(P<0.01), the statistics result of the expression of VEGF was higher in middle-dosage group(P<0.05). Compared with DM model group, the statistics result of the expression of VEGF were lower in high-dosage group, middle-dosage group and positive drug control group(P<0.01).2. Compared with normal control group, the statistics result of the expression of PEDF were lower in DM model group and middle-dosage group(P<0.01), the statistics result of the expression of PEDF was lower in low-dosage group(P<0.05). Compared with DM model group, the statistics result of the expression of PEDF was higher in high-dosage group(P<0.01), the statistics result of the expression of PEDF was higher in positive drug control group(P<0.05).3. Compared with normal control group, the statistics result of the expression of IGF-1 were higher in DM model group, middle-dosage group and low-dosage group(P<0.01). Compared with DM model group, the statistics result of the expression of IGF-1 was lower in high-dosage group (P<0.05).Conclusion:The method of activing blood,Tonifying qi and nourishing yin can effect expressions of retinal angiogenesis regulation factors of diabetes mice and improve the retinal structure and function.The method was embodied by Compound Xueshuantong Capsule. |
