Rheumatoid arthritis( RA) is a common systemic autoimmune disease characterized by symmetrical arthritis and abnormal proliferation of synovial, which is highly teratogenic. The clinical symptoms of RA is similar to other joint diseases, It is often misdiagnosed. Relevant studies show that RA is reversible in the early onset,if diagnosis and treatment timely, not only can control the disease development, but also can reduce the damage of bone and joint and improve prognosis, Therefore, early detection and diagnosis whether suffer from RA is very important.The reagent which can detect RA early and correctly needs to have both specificity and sensitivity, this paper from the protein-ligand specificity combined perspective, through the target protein of retrieval technology, high-throughput virtual screening ligands with high sensitivity, and provide a lead for the development of RA detection reagent.1, Starting from key inflammatory factor TNF-α which mediate rheumatoid arthritis, diagnosis is conducted based on the characteristics of target TNFA_HUMAN, define the target protein binding site by PFSC, high-throughput virtual screening the PDB to find out small molecule 307 and NAG which has high sensitivity, and they can used as a probe to detect rheumatoid arthritis.2, Large amounts of data is accumulated in development and application of drug,from the perspective of drug protein-ligand binding specificity, take the drugs approved by the FDA into consideration, high-throughput screening of the Protein Data Base to find small molecule ligands with high sensitivity with TNFA_HUMAN,we can know that the drug ligand CLQ, OTX has poor sensitivity, they are not suitable for used as a probe to detect rheumatoid arthritis.3, From the point of serological markers of RA——rheumatoid factor, making use of PFSC to analyze the rheumatoid factor protein-ligand binding region, screen the ligands to obtain the NAG which is specificity and high sensitivity, and NAG can be used as a probe to detect rheumatoid arthritis.The methods can be extended to the development of detection reagents andtreatment drugs for other critical diseases, which has a wide applications.
