In order to investigate the effect of Shandan Sphallerocarpus gracilis polysaccharide(SGP) on the chemical hepatic injury induced by carbon tetrachloride(CCl4)in mice.The SGP extracted from water extraction and alcohol sinking was gavaged to observe the effects of them on the chronic or acute hepatic injury induced by CCl4 with intraperitoneally injected.And the mechanism of these effects were analyzed and discussed.Hope for our works could be helpful in the prevention and treatment of alcoholic hepatic injury,and promote the development and utilization of SGP.Methods1.40 mice were divided into five groups,including the nature control group,the experimental control group,and the SGP groups.The mice in SGP groups were administered with SGP at the doses of 12.5,25,37.5g/L 0.2m L respectively,the control groups were gavaged with the same amount of saline at the same time.Then,the mice in the experimental control group and the SGP groups were injected 0.2m L CCl4 solution(1%CCl4 olive oil) after 7 days,intraperitoneally.Executed after 16 h,Weighed and recorded the body weight and liver weight in mice.The morphological changes for hepatic tissues were observed under the microscopy,the activities of plasma ALT,AST,ALP and SOD,GSH,as well as content of MDA in liver were determined by colorimetry,the expression change of caspase-3,TNF-α,Bax and TGF-β1 in liver tissue was determined by immunohistochemical method.2.Total 40 mice were used in the present study and divided into 5 groups,including the nature control group,the experimental control group,and the SGP groups.The mice in the experimental control group and the SGP groups were injected 0.2ml CCl4solution(0.2% CCl4 olive oil) per 3 days,intraperitoneally.The mice in SGP groups were administered with SGP at the doses of 12.5,25,37.5g/L 0.2m L,respectively,followed by 0.2m L(0.2% CCl4 olive oil) injection also.The body weight and liver weight in mice were be weighed and recorded.The morphological changes for hepatic tissues were observed under microscopy,the activities of plasma ALT,AST,ALP and SOD,GSH,as well as content of MDA in liver were determined by colorimetry,the expression change of caspase-3,TNF-α,Bax and TGF-β1 in liver tissue was determined by immunohistochemical method.Results1.In the acute experiment:The activities of ALT,AST and ALP in plasma were significantly increased in the experimental control group,compared with the nature control group(P<0.01),while the activity of SOD and GSH in liver tissue were decreased obviously(P<0.01),and the content of MDA were increased significantly(P<0.01),at the same time, the weight of mice were normal,the liver was obviously swelling(P < 0.01),also the livers cell were inflammatory necrosis and cavitation, The activities for Caspase-3, TNF-α, Bax and TGF-β1 immunohistochamical positive staining in hepatic tissues were significantly increased in the experimental control group(P<0.01).2.As compared with experimental control group,the activity of SOD and GSH in liver tissue were increased significantly(P<0.05 or P<0.01),However,the activity of plasma ALT,AST and ALP in the SGP groups were significantly decreased(P<0.01),and the content of MDA were also significantly decreased(P<0.01),the weight was increased,the liver was looked normal,hepatic cord was clear,inflammatory necrosis were decreased,liver celluar stucture was clear,the positive expression of caspase-3, TNF-α, Bax and TGF-β1 protein were decreased significantly,compared with the experimental control group(P<0.01).3.In the chronically experiment:The activities of plasma ALT,AST and ALP were significantly increased in the experimental control group,compared with the nature control group(P<0.01),while the activity of SOD and GSH in liver tissue were decreased obviously(P<0.01),and the content of MDA were increased significantly(P<0.01).at the same time, the livers tissue structure in experimental control group was blur,liver cells were necrosis and seriously vacuolation,The activities of caspase-3,TNF-α,Bax and TGF-β1 immunohistochamical positive staining in hepatic tissues were significantly increased in the experimental control group(P<0.01).4.However,the activity of plasma ALT,AST and ALP in the SGP groups were significantly decreased(P<0.01),and the content of MDA were also significantly decreased(P<0.01),on the contrary,the activity of SOD and GSH in liver tissue were increased significantly(P<0.01).on the organization structure, the liverstructure were renewed normal,inflammatory necrosis were decreased, and liver celluar stucture was clear,the positive expression of caspase-3,TNF-α,Bax and TGF-β1 protein were decreased significantly, compared with the experimental control group(P<0.01).Conclusion1.SGP influenced the body weight,liver weight and liver coefficient of mice caused by CCl4,which could increase body weight and reduce liver swelling degree in mice;2.SGP could increase the stability of liver cell membranes,to keep the activity of ALT,AST and ALP in plasma increasing,and had a role in regulating the liver metabolis;3.SGP could eliminate effectively oxygen free radical in liver tissue damaged by CCl4,the activities of SOD and GSH could be improved while the content of MDA be decreased.4.SGP may promote cell proliferation and repair damaged by CCl4,and had a role in protecting the liver morphology and organization structure,and have a dose effect within a certain range.5.SGP could prevent apoptosis cascade reaction,reduce the apoptosis expression of Caspase-3,Bax protein in damaged liver tissue.6.SGP may inhibit the inflammation in damaged liver,reduce the positive expression of TNF-α protein.7.SGP could reduce the expression of TGF-β1 protein in liver tissue,inhibit the production of extracellular matrix and deposition,prevent liver fibrosis.
