Objective:Chrysanthemi Flos,one top grade drug of "Shen Nong’s Herbal Classic",is the dried capitulum of Compositae Plants Chrysanthemum morifolium Ramat,which is a famous traditional chinese medicine and widely distributed in Hena.Chrysanthemi Flos has the function of heat-clearing and detoxicating,wind-heat disperseing,liver-pacifying and vision-improving,which is commonly used to treat sore abscess,deep-rooted boil,lung abscess,discomfort in the throat,influenza and so on.Modern pharmacological research also shows that chrysanthemi Flos has anti-inflammatory activity.Professor Toshihiro Akihisa group from Nihon University has found that the triterpene diols and triols isolated from the non-sapoifiablelipid(NSL)fraction of the methanol extract of the edible chrysanthemum ligulate flowers possess remark-able anti-inflammatory activity.Chrysanthemi Flos is the city flower of kaifeng,the ancient capital of seven dynasties,and is as well as GI protection products.Chrysanthemi Flos cultivated in Kai Feng has long been used for ornamental and edible,however investigation on its active ingredient and pharmacological action are rarely reported.Acute lung injury(ALI)mostly induced by endotoxin from pathogenic microorganisms metabolism,which is similar to heat toxin syndrome of TCM.In this eassy,we try to investigate the protective effect of Huangfeiwu of Chrysanthemi Flos cultivated in Kai Feng,and study the anti-inflamatory mechanisms of Huangfeiwu by the model of actue lung injure induced by lipopolysaccharide in mice.Methods:Research on Huangfeiwu of Chrysanthemi Flos cultivated in Kai Feng,it is efficient way for the extract preparetion when Huangfeiwu is heating reflux by 20 times of 95%ethanol,then purified by chloroform extraction.The mice ALI model was established by lipopolysaccharide(LPS)intraperitoneal injection.Mice were respectively killed at 3,6,and 24h time point after injection of LPS.White blood cell(WBC)count in blood,the ratio of wet lung to dry lung weight(W/D),lung index(LI)and left lung dry weight/body weight ratio(D/B)was measured.the changes of TNF-α,IL-6,IL-10,TGF-β1 in lung tissue and serum,T-AOC activity and MDA content in lung tissue was determined respectively.Optical microscope was applied to observe lung morphological changes in mice.The content of total flavonoid,total triterpenoid and taraxasterol were determined by UV-vis spectrophotometry and high performance liquid chromatography to primarily discuss the anti-inflammatory substance foundation of the Huangfeiwu extract.Results:Compared with the model group,the high dose group of Huangfeiwu significantly decreased the white blood cell(WBC)count,lymphocyte(LY)count,granulocyte(GR)count;as well as descreased the value of W/D,the lung index,the value of D/B;T-AOC activity and MDA content in lung tissue.Meanwhile,the extract(200 mg/kg)of Huangfeiwu also decreased the level of inflammatory cytokine TNF-a,IL-6,and increased the level of anti-inflammatory cytokine IL-10,TGF-β1.The content of total flavonoids,total triterpenoids and taraxasterol in the extract is up to 30.3%,0.86%and 1.315%respectively.Conclusion:Huangfeiwu extract could decrease the level of pro-inflammatory cytokines TNF-a,IL-6,and increase the level of anti-inflammatory cytokine IL-10,TGF-β1 to keep the balance between anti-linflammato factors and pro-inflammatoryf actors.Besides,it could enhance T-AOC activity in lung tissue,reduce MDA content in lung tissue,and inhibit oxygen free radicals elevation.Thus,the extract has the protective effect against ALT.The total triterpenoid may be the anti-inflammatory substance foundation of Huangfeiwu extract. |
