1.Myeloid leukemia factor functions in anti-WSSV immune reaction of kuruma shrimp,Marsupenaeus japonicusMyeloid leukemia factor(MLF)plays an important role in development,cell cycle,myeloid differentiation,and regulates the RUNX transcription factors.However,the function of MLF in immunity is still unclear.In this study,an MLF was identified and characterized in kuruma shrimp Marsupenaeus japonicus,and named as MjMLF.The full-length cDNA of MjMLF contained 1111 nucleotides,which had an opening reading frame of 816 bp encoding a protein of 272 amino acids with an MLF 1-interacting protein domain.MjMLF could be ubiquitously detected in different tissues of shrimp at the transcriptional level.The expression pattern analysis showed that MjMLF could be upregulated in shrimp hemocytes and hepatopancreas after white spot syndrome virus challenge.The RNA interference and protein injection assay showed that MjMLF could inhibit WSSV replication in vivo.Flow cytometry assay showed that MjMLV could induce hemocytes apoptosis which functioned in the shrimp antiviral reaction.All the results suggested that MjMLV played an important role in the antiviral immune reaction of kuruma shrimp.The research indicated that MjMLF might function as a novel regulator to inhibit WSSV replication in shrimp.2.Myeloid leukemia factor participates in the innate immune defense against Vibro anguillarum of kuruma shrimp M.japonicusMyeloid leukemia factor(MLF)plays an important role in the development of Drosophila.And it regulates cell cycle via p53-depended manner in human.Some researches focus on MLF’s translocation between cytoplasm and nucleus in mouse.It is reported that MLF could inhibit white spot syndrome virus(WSSV)replication in shrimp.However,the function and mechanism of MLF in bacteria immunology are still elusive.In this study,we found MjMLF could be upregulated by Vibro anguillarum challenge in hemocytes,gills,and intestine of kuruma shrimp.MjMLF participated in bacteria clearance by RNAi assay,and it was beneficial to shrimp survival in V.anguillarum infection and able to bind to Mj 14-3-3.Some antimicrobial peptide genes could be regulated by MjMLF.These results indicated that MjMLF might participate in the shrimp immune reaction against bacteria.
