| Marsdenia tenacissima(Roxb.)Wight et Am.is a perennial climber belonging to the Marsdenia genus in the Asclepiadaceae family,which is used in Chinese folk medicine for the treatment of tracheitis,asthma,tonsillitis,cystitis,cancer,and pharyngitis.Clinical studies have shown that the aqueous extractions of M.tenacissima are beneficial for treating patients with various cancers.So there is a rapidly increasing demand of the market which is made up of injection and granules that are made from M.tenacissima,but at present all resources of M.tenacissima are in the wild,and there are some problems such as less seed,long growing season and so on.So it is necessary to investigate the habitats and study the seed to improve the quality of M.tenacissima.Excellent germplasm is the basis and key to ensure the quality of Chinese medicine material.It is important to study the characteristics of seed,the quality inspection methods of seed,and how to improve the germination rate under stress.This paper studies the characteristics of seed,seed priming technology which improves seed resistance,the quality classification stand of the seed of M.tenacissima from different population.The results are as follows:1.The results showed that Marsdenia tenacissima seeds can germinate in different ground substances especially in sandy soil,the effect of soaking time on seeds germination is not significant,the seeds could germinate well during 25~30 ℃.Based in appropriate conditions,the effect of illumination is not significant but the illumination should be the key factor under inappropriate temperature.The effect of acidic soil is lighter than alkaline soil.The optimal germination condition of Marsdenia tenacissima seeds is 24 h of marinating at the temperature of 30 ℃ with sandy soil which pH is 7.0~7.5 under illumination.2.Referring to the International Seed Testing Rules made up by ISTA and Rules for agricultural seed testing issued by China,the seed quality of M.tenacissima from different collection was measured.The samples weight of M.tenacissima were at least 900 g for purity analysis and 90 g for testing.The weight of 1000 seeds was determined by using 500-seed method.The water content of the seeds was determined under the higher temperature(133±2)℃ for 6 hours.After soaking in distilled water for 24 h,the seeds were cultured in wet sand at 30 ℃ for 1~8 days under illumination for germination testing.Seed viability was tested by TTC method.3.To offer the reference and method for salt damage in the cultivation of M tenacissima,in this study,high.Four different treatments,which were GA3,KNO3-KH2PO4,PEG-6000,NaCl,combined with ANOVA were applied to test the performance of germination energy,germination percentage,germination index,MDA,SOD,and CAT.The results showed the seed germination was obviously inhibited under salt stress and the soaked seeds with different priming materials could alleviate the damage of salt stress.Under these treatments,the activities of SOD,CAT the content of soluble protein were significantly increased,While the content of MDA was significantly decreased.The maximum index was obtained when 1.20%KNO3-KH2PO4 treatmented,with the germination percentage increasing from 52.67%to 87.33%and the activity of SOD increasing from 138.01 to 219.44 respectively.Compared with the treatment of 1.20%KNO3-KH2PO4,the germination percentage of treating with 300 mg·L-1 GA3 was increased from 52.67%to 80.67%,while the activity of SOD was increased from 138.01 to 444.61.4.The results showed the seed germination was obviously inhibited under drought stress and the soaked seeds with different priming materials could alleviate the damage of drought stress.Under these treatments,the activities of SOD,CAT the content of soluble protein were significantly increased,While the content of MDA was significantly decreased.The maximum index was obtained when 1.60%KNO3-KH2PO4 treatmented,with the germination percentage increasing from 47.33%to 90.67%and the activity of SOD increasing from 94.92 to 186.70 respectively.Compared with the treatment of 1.60%KNO3-KH2PO4,the germination percentage of treating with 300 mg · L-1 GA3 was increased from 47.33%to 89.33%,while the activity of SOD was increased from 94.92 to 179.22. |
