Study On Pharmacology And Clinical Therapeutic Efficacy Of Compound Zhongjiefeng Particle

Objective:The Compound Zhongjiefeng Particle is the new pure traditional Chinese drug jointly developed by Guangxi University and Guangxi Big Dipper Animal Health Products Co.,Ltd.Previous studies have revealed that the drug has the effects of detoxification,clearing the lungs phlegm,reducing swelling and removing stasis,relieving cough and asthma and reducing uric acid.In this study,series of pharmacological and clinical researches were conducted to evaluate the effects of Compound Zhongjiefeng Particle on preventing and treating chicken hyperuricemia cough and asthma so as to provide a scientific basis for rational use of this drug.Methods:(1)An acute hyperuricemia Artificial model in mice was made by intraperitoneal injection of potassium oxonate.The protective effects of different doses of Compound Zhongjiefeng Particle in hyperuricemia mice were investigated by means of physiological and biochemical indexes,organ index,kidney tissue pathological slices;(2)A hyperuricemia Artificial model in chicken was induced by feeding with food containing both high calcium(4%)and protein(28%)to evaluate the prevention and treatment effects of Compound Zhongjiefeng Particle.Prevention trials:The chicken with hyperuricemia were given at different doses of the Compound Zhongjiefeng Particle at the same time.The prevention effect was evaluated by measuring the values of serum uric acid and other biochemical indicators,combining clinical symptoms,morbidity,cure rate and effective rate.Treatment trials:The chicken with artificially induced hyperuricemia were treated with different doses of Compound Zhongjiefeng Particle.The treatment effect was evaluated by measuring the values of serum uric acid and other biochemical indicators,combining clinical symptoms,morbidity,cure rate and effective rate.(3)A cough and gasp pathology model in chicken was artificially induced by IBV.One group was given the Compound Zhongjiefeng Particle mixed in drink three days before challenged,and the other groups were given at different doses of Compound Zhongjiefeng Particle after cilnical signs appeard.The prevention and treatment effects of the Compound Zhongjiefeng Particle in chicken with cough and gasp was observed by measuring biochemical indexes,mortality rate,the total effective rate,relative weight gain rate,immune organ index of the chickens each group.Results:(1)Compared with the Normal Control,the levels of serum uric acid in Positive Control were significantly higher(P<0.01),indicating that the application of the potassium oxonate can be used to make model;Compared with the Positive Control,the levels of serum uric acid in treatment groups were significantly lower(p<0.01),showing that Compound Zhongjiefeng Particle and control drug played an important role in decreasing uric acid;The activity of xanthine oxidase(XOD)in the liver homogenate of in high-dose and medium-dose group were significantly lower(P<0.05).The result proved that the role of the drug in descending uric acid may be associated with activity XOD.(2)The chickens infected with IBV appeared clinical symptoms,including loss of appetite,listlessness,hair loss,diarrhea,feces containing a large number of white matter,together with value of serum uric acid over than 416 μmol/L.Prevention trials:the morbidity of animals treated with the drug was far less than the infection control.The morbidities of high-dose,medium-dose,low-dose groups and Jitongfengxiaosan control group were 3.3%,13.3%,30%and 16.6%respectively;The total efficiency were 96.7%,86.6%,70.7%and 83.2%respectively.Treatment trials:the total effective rates of high-dose,medium-dose,low-dose groups and Jitongfengxiaosan control group were 99%,93%,93%and 76%.(3)The EID50 of IBV-M41 was 10-6’68/0.2ml;The chicken showed the typical clinical symptoms of cough and asthma 4d after challenged.The specific gene band of IBV was detected with PCR at 740 bp.The efficiency of all treatment group as follow:the prevention group,high-dose group and medium-dose group were 70%,the low-dose group was 60%,the Maxinshigansan control group was 50%.Conclusion:(1)The Compound Zhongjiefeng Particle had an effect in descending uric acid,in a dose dependent manner and its mechanism was likely related to the inhibition of hepatic xanthine oxidase activity.(2)The Compound Zhongjiefeng Particle had effects in preventing and treating chicken hyperuricemia.The recommended prevention program was 0.6 g per kg body weight each day in drink for 7 days;The recommended treatment program was 1.2 g per kg body weight each day in drink for 5 days.(3)The Compound Zhongjiefeng Particle had effects in preventing and treating of chicken cough and asthma.The recommended prevention program was 1.2 g per kg body weight each day in drink for 5 days;The recommended treatment program was 2.4 g per kg body weight each day in drink for 5 days.
