| 40 different wild letinous edodes strains and 8 different conventional letinous edodes strains were collected from the introduction as experimental material.The classified and identification,evaluation and genetic diversity research was studied by using the esterase isoenzyme clustering analysis,ISSR molecular marker technology and fruiting test.(1)15 enzyme bands with different relative mobility were detected by the esterase isozyme zymogram determination of 40 wild letinous edodes strains.Through the cluster analysis,the results showed that,when the genetic similarity coefficient was about 71%,the zymogram types of the 40 wild letinous edodes strains were divided into seven categories,the 40 test strains can be divided into seven categories.8 enzyme bands with different relative mobility were detected by the esterase isozyme zymogram determination of 8 conventional letinous edodes strains.Through the cluster analysis,the results showed that,when the genetic similarity coefficient was about 86%,the zymogram types of the 8 test conventional letinous edodes strains were divided into four categories,the 8 test conventional strains can be divided into four categories.(2)ISSR molecular marker experiments of 40 wild letinous edodes strains showed that 175 polymorpHic loci between 200 bp and 4000 bp molecular weight could be obtainedused through gel electropHoresis used by 6 primers respectively.Through the cluster analysis,the results showed that,when the genetic similarity coefficient was about 76%,the 40 test strains can be divided into ten categories.8 conventional letinous edodes strains were amplified by ISSR with 5 selected primers,the results showed that 49 polymorpHic loci were amplified.Through the cluster analysis,the results showed that,when the genetic similarity coefficient was about 88%,the 8 test conventional strains can be divided into four categories.(3)based on two kinds of different cultivation patterns(plant type,plant type)mushroom contrast test,it is concluded that to plant type is relatively suitable for mushroom cultivation method for fruit intercropping bacteria.(4)The fruiting test of 40 wild strains and 6 conventional strains showed that thewild letinous edodes strain “Le49”,“Le70” and the conventional letinous edodes strain “60” were selected as the suitable strains for Interplanting between fruit and fungi. |
