In this paper,the establishment of Radix Tetrastigme tissue culture and rapid proliferation system were studied.In this system,we induced the axillary buds to obtain the aseptic seedlings,and we compared the effects of different phytohormone combinations and concentrations on the culture of axillary bud induction,axillary bud proliferation,rooting and callus induction.Total flavonoids were obtained by ultrasonic-assisted extraction,and its contents were determined by UV-Vis Spectrophotometry.Polysaccharides were obtained by hot water extraction,and its contents were determined with Phenol vitriol colorimetry by UV-Vis Spectrophotometry.Results showed that:1.Disinfecting methods of explant:The most effective disinfection method-the axillary buds were immersed with 75%alcohol for 5 minutes,sterilized with 0.1%mercuric chloride(plus a few drops of Tween)for 10 minutes,the success rate was the highest(53.92%).2.Axillary bud induction:The most effective culture medium for axillary bud induction was MS+2.0mg/L 6-BA+0.2mg/L NAA,and the inducing rate was 51.67%.3.Axillary bud proliferation:The most effective culture medium for axillary bud multiplication was MS+2.0mg/L 6-BA+2.0mg/L NAA,and the multiplication ratio reached 3.1.4.Rooting:The most effective culture medium for rooting was 1/2MS+1.0mg/L IBA +0.1 mg/L NAA,the rooting rate reached 81.6%.5.Callus induction from leaf blade:The most effective culture medium for callus induction from leaf blade was 2/3MS+2.0mg/L 6-BA+2.0mg/L NAA,the rate of callus induction reached 76.3%.6.The total flavonoids content of callus was closed to the total flavonoids content of the same roots,and the polysaccharide content of callus was even higher than the polysaccharide content of the same roots.
