| The Glycyrrhiza uralensis Fisch was researched in this study.First,the salt accumulation characteristic and salt secretion characteristic of Glycyrrhiza uralensis are explored,Glycyrrhiza uralensis seedlings were cultured with the complete Hoagland nutrient solution containing 0,80,160 and 320 mmol·L-1 NaCl for 21 days respectively,then,the content of K+,Ca2+ and Na+ secreted from the leafs were measured by ICP-AES,in addition,two-year-old Glycyrrhiza uralensis grew in heavy saline-alkali soil were collected,the content of K+,Na+,Ga2 + and Mg2 + in different parts(roots,rhizomes,stems,old leaves and young leaves)of Glycyrrhiza uralensis were measured by ICP-AES.Second,this study explore the salt ion micro area distribution of different organ(roots,rhizomes,stems and leaves)of Glycyrrhiza uralensis by Scanning Electron Microscopy and Energy Spectrometer.Third,organ surface(stems,leaves,petioles,calyx,petals and ovary)of Glycyrrhiza uralensis was scaned by Scanning Electron Microscopy to find whether salt glands also exist on these organ surface;Fourth,whether salt glands is selective to secrete salt ion was explored,Glycyrrhiza uralensis seedlings were cultured with a complete Hoagland nutrient solution containing 100mmol·L-1 NaCl,KCl,CaCl,MgCl respectively,then the content of salt ion secreted from the leafs were measured by ICP-AES.The aim for this study is to provide reference for salt tolerance theory.The main results:1 More salt ions secreted from leafs of Glycyrrhiza uralensis with the increased concentration of NaCl,the content of Na+ secreted from leafs was more than Ca2+ and K+;In high salt habitats,the young leaves is the main part to accumulate K+,the old leaves is the main part to accumulate Ca2+.The content of Na+ in every part of Glycyrrhiza uralensis is very low and it is obvious characteristic that Glycyrrhiza uralensis reject accumulating Na+.2 The results of salt ion micro area distribution shows that Na+ mainly accumulate in epidermis of roots and rhizome of Glycyrrhiza uralensis;As for stem,Na+ mainly accumulate in the cortex;What more,Na+ also mainly exist in the epidermis of leafs.3 Salt glands not only be observed on the surface of leafs but also on the stems,petioles and calyx of Glycyrrhi za uralensi s,the result prove Glycyrrhiza uralensis have more secretory pathway than other plants.4 Glycyrrhiza uralensis seedlings was respectively stress by 100mmol/L KCl,NaCl,GaCl2 and MgCl2 and the result shows that the most content of salt ion secreted from leafs respectively are K+、Na+、Ca2+ and Mg2+,salt gland on the leafs is non-selective to secrete salt ions. |
