| Endophytic fungi of drunken horse grass(Achnatherum inebrians)was Epichloe gansuensis form mutualistic symbionts after infection.Epichlo? endophyte symbiotic Achnatherum inebrians produce two important ergot,ergonovine and ergine,which make the symbionts have certain toxicity of domestic animals and insects.Drunken horse grass endophytic fungi seedling were observed as the research object,mainly including drunken horse grass growth and the change of ergot alkaliold contents under different temperature,p H and light processing,detection method of ergonovine and ergine is established by using high performance liquid chromatography(HPLC).The main results are as follows:1.The standard curve of lolitrem B,peramine,homoperamine,ergine,ergonovine and ergovaline alkaloids wereestablished by high performance liquid chromatography(HPLC).In order to follow-up for qualitative and quantitative determination ofthe various types of alkaloids.2.By investigating the effects of different temperatures(8/5℃,15/12℃,22/15℃,28/25℃,35/28℃)on variations of growth and ergot alkaloids concentrations in Achnatherum inebrians seedling with Epichloёsp.,it was found thata short periods of high temperature is good for two ergot alkaloidsaccumulation,on the contrary,a long periods of high temperature is not.As treat time goes on,the production of two ergot alkaloidsaccumulation is highest under moderate temperature,and physiological indexs also better than other treat,so the optimum of symbionts seedling growth,the accumulation of ergine and ergonovine treatment temperature is 22/15℃.3.By recording and measuring plant height,root length,biomass,chlorophyll,soluble sugar,ergine and ergonovine contents under different p H(3,5,7,9 and 11),it was found that symbiotic seedlings grow best under neutral conditions and under alkaline conditions grow well more than under acidic conditions.However,the accumulation of ergine and ergonovine had peak values under strong alkaline conditions.Comprehensive growth and ergot alkliold production,the symbionts can be used in the cultivation of saline-alkali land greening.4.By recording and measuring plant height,root length,biomass,chlorophyll,soluble sugar,ergine and ergonovine contents under different photoperiod(8 h·d-1,12 h·d-1,14 h·d-1and 16h·d-1)and illumination intensity(1500 lux,2500 lux and 2500 lux),it was found that symbiotic seedlings grow best under 16 h·d-1and 3500 lux,and the most suitable conditionfor the accumulation of ergonovineand ergine is12 h·d-1 and 2500 lux. |
