| The bleeding sap refers to xylem sap which comes from the wound of plant.Bleeding period is an important stage before the germination and the generation which be associated with the activity of root.On the basis of previous study,this study focused on the physiology and changes of important genes involved in grapevine floral development with’Wink’ as research material.This research made a further discussion on the influence on the flowering progress of bleeding sap,in order to make a further analysis on the functions of plant sap.The main research results of this study are as follow:1.Bleeding sap postponed the phenophase of grapevine,which resulted in the delay of budding and florescence.Bleeding sap also resulted in the decrease of the rate of germination.The sap put off the growth of new shoot,decreased area of leaves and brought the length of inflorescence down.2.The generation of sap stimulated the stress reaction of grapevine.By analyzing the osmotic regulating substances and stress production,the content of them increased with the bleeding.In addition,the activity of protective enzyme had an obvious rise.3.This study did analysis on the relative expression of several important genes in grapevine floral development by quantitative PCR.The results showed that the relative expression of floral positive regulating genes was lower than that of the control group,which illustrated that bleeding inhibited the expression of these genes.Expression of VvLFY between treatment group and control group was the greatest different.By analysis on the upstream regulating genes of different florescence pathway,it found out that VvSPL10,which related to the gibberellin pathway had the corresponding difference.Combined with the determination of GA3 in sap in our lab,it speculated that the effects of the bleeding sap on the florescence may be influenced by the gibberellin pathway or nutritional pathway,the other 3 pathways as supplement. |
