Goose is favored by customers due to its relative high nutritive value with low fat and high unsaturated fatty acid.It isreported that the production of muscle is closely related to the amount and growth of muscle fiber cells.Skeletal muscle fiber is formed by muscle cells proliferation and differentiation,the proliferation and differentiation of muscle cell is controlled by multiple factors in a double-edged way.MRFs participate in the progress of muscle growth,and control the proliferation and differentiation of muscle cells,which play an important role in the process of skeletal muscle differentiation during the embryonic stage and the early stage of hatch.Pax3 and Pax7 are considered to guide the formation of various processes during the growth and development of muscle fiber.Recently,Tmem8 c is found as a muscle-specific membrane protein,whichcontrols myoblast fusion.However,few researches were reported on these genes during the embryonic and muscle development in goose.In this study,we chosed goose as material.HE staining wasused to observe the growth of breast and leg muscle during embryonic period,RACE was takento obtain the full length of Tmem8 c,QRTPCRwas done to analyze the expression of each gene,and LRGassay to predict the candidate miRNA of Tmem8 c.Our results revealed(1)the results of HE staining showed that the breast muscle and leg muscle hadexperienced from myoblasts to multinucleated myotubes to muscle fibers,and leg muscle was developed earlier than breast leg.(2)QRT-PCR indicated that MRFs,Pax3 and Pax7 were all relative to the development of goose skeletal muscle with different expression patterns.MRFs and Pax7 showed the peak expression during the period of E11 to E15,and the highest expression of Pax3 appeared at the beginning of hatch followed by the decline expression,which suggested that the expression patterns of these genes were highly matched with their function during muscle development.(3)The Tmem8 c gene of goose had high homology with other species,and had some conserved features.(4)Tmem8c had a peak expression pattern,reaching its highest expression levels in stage E15 in LM and E19 in BM,and then droppedtransiently in E23(P<0.05).(5)By LRG assay and correlation analysis,it was found that miR-16-1 might be a potential miRNA ofTmem8 c.
