| The first human autosomal gene that has been found to play an important role in the maintenance of ovarian function and ovarian development is the FOXL2 gene,also was the first discovered in vertebrates ovarian differentiation sexual dimorphism state marker gene for the first discovered ovarian differentiation.With the deepening of the related research in human,it was found that the FOXL2 gene was expressed specifically in human eyelid,embryo and adult granulosa cells,relating to the sex determination,POF,infertility,ovarian tumor and small palpebral fissure syndrome and other diseases.FOXL2 gene plays an important role in the inhibition of follicular apoptosis and the promotion of ovarian reserve function,its normal expression is very important for egg laying performance and ovary development.FOXL2 gene plays an important role in the inhibition of follicular apoptosis and the promotion of ovarian reserve function,its normal expression is very important for egg laying performance and ovary development.But the expression of FOXL2 gene in different tissues and organs of laying hens and there was no report on the correlation between the different stages of ovarian development.This study first to observe the development and histologic structure of the ovary ovaries at different times by HE staining,and through the statistical analysis of the follicles’ s number to study the specific development of the ovarian follicles.According to the FOXL2 sequence of GenBank gene included red jungle fowl(accession number: JF708868),analysis its conservative sequence and the enzyme site by bioinformatics,design and synthesis of 3 pairs of primers by Primmer5.0.Through agarose gel electrophoresis experiment screen out the optimal primer.Collection 30-day old,60-day old,90-day old,120-day old,160-day old,220-day old,330-day old,480-day old that total of 8 periods of Hy-Line Brown’s hypothalamus,eyelid,muscle,heart,thymus,liver,spleen and kidney tissue samples,to conduct qRT-PCR experiment.Comparison of the relative expression of FOXL2 gene in different stages of ovary,analyze the FOXL2 gene’s relationship with ovariandevelopment and egg laying performance;comparison of the relative expression of FOXL2 gene in different periods and different tissues,analysis the correlation between FOXL2 gene and the development of laying hens.The results of HE staining and statistical analysis showed that the size and weight of the 220-day old ovaries are the largest,later the ovaries gradually atrophy and losing weight;With the increasing of age,the number of total follicles decreased gradually,and the number of primary follicles increased first and then decreased,the60-day old’s primary follicle number is the highest,and then gradually decreases;the number of secondary follicles of 120-day old was the highest,and significantly higher than the other laying hens;from the 30-day old,the number of follicles is increasing with the increase of date.The results of the qRT-PCR experiment showed that the relative expression of the FOXL2 gene in the ovaries of the 120-day chicken ovary was higher than that of the other days.The expression of FOXL2 was correlated with the development of the layer ovary,the relative expression of the FOXL2 gene in different tissues of laying hens was accompanied by the development of organ and tissue,it shows that the FOXL2 gene had some relationship with the development of other tissues.This study is helpful to further understand the role of the FOXL2 gene in the regulation of ovarian development and laying performance,this research laid the foundation for the further study on the biological characteristics and function of the gene. |
