Stalk Chinese Cabbage is a cruciferous Brassica Vegetable Crop.In order to play its heterosis,the use of male sterility is one of the most desirable methods.In this study,the GMS material of Stalk Chinese Cabbage were selected from several lines by our research group.In this study,in order to make better use of this precious male infertility genetic resources,explore the advantages of the male sterile line of Stalk Chinese Cabbage,and apply it to breeding practice of Stalk Chinese Cabbage as soon as possible,we conducted the primary mapping using SSR molecular marker technology on the basis of genetic analysis of the sterile gene.The results are as follows:In this study,a small amount of sterile plants found in the original materials sib mating with different fertile lines.We chose the offspring of fertile separation of the group to continue sib mating and made fertility and cross identification of progeny.We get 3 Single-gene recessive nuclear male sterilities dual purpose lines ’Xiangtai SN-1’,with the proportion of sterile lines to fertile plants is maintained at 1:1 and stable sterility.The male sterile lines of Stalk Chinese Cabbage were used as the female parent,and the homozygous recessive genotype msms Chinese cabbage of our laboratory as the male parent,and the offspring were all fertile.Therefore,the sterile genes of Chinese cabbage were found to be different from the Stalk Chinese Cabbage sterile genes found in our laboratory.In this study,we used the Xiangtai SN-1 sterile plant as the female parent and Chinese cabbage DH lines ’FT’ as the male parents to build a F2 mapping population which include 7656 plants.The fertility of 7,656 strains of F2 population was investigated.Among them,5782 fertile plants and 1874 sterile strains were found.The fertility phenotype conformed to the 3:1 separation law.We analyzed SSR molecular markers and conducted the primary mapping using 1874 male sterile plants in F2 population.We selected the polymorphism in the parents using 700 SSR markers which explored by our research group.Only 4 markers show stable and close linkage relationship with the male sterile gene.They are cnu_m561a,nia_m032a,nia_m068a and nia_m071a which all come from Chinese cabbage A01 linkage group.54 recombinant plants were found when detected the marker nia_m068a in the fine mapping group with the genetic distanct of 1.441 cM.
