Plant virus disease is a serious diseases on agricultural production,which has brought severe losses to the output of crops.In recent years,Microbial source antiviral substances by its unique advantage have become a research focus in the screening of antiviral substance.We take Nicotiana glutinosa、Nicotiana tabacum and Tobacco mosaic virus(TMV)as the host material and experiment virius,combing with the active tracking and modern separation technology,the Anti-TMV Active componets of endogenous tobacco were researched.The main results were shown as follows:(1)By half leaf spot and leaf discs of tobacco plants,suspended screening method of Yunyan 203,Nicotiana glutinosa,Nicotiana tabacum three kinds of tobacco plant,root,stem and leaf of tobacco,methanol extracts were determined from anti TMV activity.The results showed that three kinds of flue-cured tobacco in the whole plant,root,stem and leaf of tobacco and methanol extracts inhibited the branch activity on TMV in vitro passivation,protection and treatment,the best root activity,compared with the whole plant inhibitory rate of TMV increase more;stems,leaves and branches.The results showed that the three kinds of tobacco root extracts showed significant dose effect in vivo by Nicotiana tabacum.(2)The half leaf spot of Yunyan 203,Nicotiana glutinosa,Nicotiana tabacum on activity of anti TMV three kinds of flue-cured tobacco roots of different polar solvent extracts,found three kinds of flue-cured tobacco rootsof ethyl acetate part passivation of TMV,protective effect and therapeutic effect of strong.(3)Nicotiana tabacum on the root of the ethyl acetate extract by silica gel cross section,dichloromethane: methanol 9:1(Fr.2)activity further enhanced inhibition of passivation,protective and therapeutic effects on TMV infection rate of 93.06%,85.03% and 71.54% respectively;further draw on Fr.2 section is divided into three parts,the three part of the TMV infection showed strong activity.The activity of Fr.2,Fr.2-1,Fr.2-2 and Fr.2-3 was similar to that of the other components of the root extracts after further validation.(4)This paper uses a variety of chromatographic techniques(silsca gel,Sephadex LH-20,ODS,HPLC,MCI)component separation of ethyl acetate parts of Nicotiana tabacum root.Structural identification of compounds were isolated by MS,NMR,IR and UV spectroscopy.Totally 40 compounds were isolated from the roots of Nicotiana tabacum,and the compounds were identified in the presence of 17 compounds.The structure types include lignans(1-2),flavonoids(17),coumarin(4-12)and benzene derivatives(3,13-16)and so on.Among them,2 new compounds(1-2).They are Spondlakin A(1),SpondlakinB(2),1-β-D-glucopyranosyl-2,6-dimethoxy-4-Propenylphenol(3),Micromelin(4),AcetylhydromicromelinA(5),DihydromicromelinA(6),DihydromicromelinB(7),Hopeyhopin(8),Nodakenin(9),Scopoletin(10),Scopolin(11),Magnolioside(12),2-Methoxy-4-(2-propenyl)phenylβ-D-glucopyranoside(13),Benzylβ-D-glucopyranoside(14),Dictamnoside A(15),2-(4-Ethylphenyl)ethanol(16),5,7-Dihydroxyl-3,8,4’-trimethoxylflovone(17).
