Swainsonine(SW),the primary toxic component of locoweed,can inhibit the activity of α-mannosidase and mannosidase II,leading to lysosomal storage.Animal absorbed with excess dosage will show weight reduction,mental depression,behavioral changes,infertility and other clinical features.Microbial degradation of SW can be used as a new approach to combat animal poisoning.Arthrobacter sp.HW08 is a swainsonine-degrading strain that is isolated from the soil.In order to reveal the degradation mechanism of swainsonine and to explore the survival strategy in harsh environments,complete genome sequence and analysis,substrate using test,antibiotic resistance test and plasmid detectionof Arthrobacter sp.HW08 were performed.Through third generations of sequencing,Arthrobacter sp.HW08 contained a circular chromosome of 5,063,552 bp with anaverage G + C content of 62.65%.A total of 4816 genes were obtained by gene annotation,and 4742 of them were protein-coding genes.After comparison with multiple databases,we found that nearly half of the genes in the genome of the strain were involved in biological metabolism.These genes are distributed in 13 amino acid metabolic pathways,14 glucose metabolism pathways and 11 branches of lipid metabolism pathways.Genomic analysis also showed that the bacteria had multiple genes involved in osmotic pressure regulation and oxidative stress protection,but in terms of metal ion resistance,only annotated to the copper and arsenaterelated genes.The results of the substrate utilization test showed that the bacteria could be treated with sodium benzoate,p-aminobenzene sulfonic acid,4-aminoantipyrine,L-proline,L-piperidine acid,1-methylpyrrolidine,N-methylpiperidine,dichlorvos and phoxim as the sole carbon source.The results of the antibiotic resistance test showed that the strain was resistant to kanamycin and streptomycin.But there is no resistance gene in the genome.In addition,we found a gene that encodes the bacitracin resistance protein BacA.Finally,by using pulsed-field gel electrophoresis,it was confirmed that any form of plasmid was not contained in Arthrobacter sp.HW08.
