Lung pathology showed swelling and hyperemia in the process of ascites syndrome. The lung is a highly vascular organ. It is closely related to the occurrence of AS that the structure and function of pulmonary vascular endothelial are disorder. In recent years, the research on the prevention and treatment of AS with traditional Chinese medicine has been widely and achieved good results. According to the previous research results, this study was designed to study the effect of compound traditional Chinese medicine on the regulation of endothelial cell proliferation and adhesion, and then to study its effect on the prevention and treatment of pulmonary hypertension syndrome in broiler chickensMethod and resultUsing multivariate methods(low-temperature, water add 0.12% NaCl,fodder add 3% lard and 4% fish meal) induced AS in AA broiler. Experimental animals were divided into six group:control group, model group, high-dose of Compound Traditional Chinese Medicine (CTCM) group(2 mL/kg·d),middle-dose group(1mL/kg·d),low-dose group(0.5mL/kg·d) and positive group(1%L-Arg).We determinate the mRNA and protein levels of lung tissue adhesion factor(VCAM1), Proliferation realate gene (EGFL7,VEGF,VEGFR2) and negative regulatory factor(SPRED1、PI3KR2) by real time PCR and ELISA. To study the prevention and effect mechanism of traditional Chinese medicine to ascites syndrome. The main content of the test and results are as follows:1.Listlessness,abdominal swelling;We can extract 200-500 mL of yellow fluid from the enlarged abdomen;The disease chicken with pericardial effusion, liver and lung congestion and swelling.2.Compared with the control group,the model group AHI wassignificantly increased(P< 0.01);Compared with the model group,hige-dose and middle-dose of CTCM group and L-Arg gruop AHI was significantly increased(P< 0.01, P< 0.05).3.Compared with the control group, the relative expression of EGFL7 mRNA in lung tissue of model group was significantly increased(P<0.01);Compared with the model group, the relative expression of EGFL7 mRNA in lung tissue in high-dose of CTCM group and L-Arg group was significantly decreased(P <0.01),the relative expression of EGFL7 mRNA of middle-dose of CTCM group on 15,25,35 days of age were significantly decreased(P<0.01),the relative expression of EGFL7 mRNA in low-dose of CTCM group was significantly decreased at 35 days of age(P<0.01).Compared with the control group, the expression of EGFL7 protein in lung tissue of model group was significantly increased(P<0.01);Compared with the model group, the expression of EGFL7 protein in of high-dose of CTCM group significantly decreased(P<0.01, P<0.05), middle-dose of CTCM group and L-Arg group were significantly decreased on 15,25,35 days of age(P<0.01, P<0.05),the expression of EGFL7 protein in of positive of CTCM group were significantly decreased on 15 days of age(P<0.01).4.Compared with the control group, the relative expression of VEGF mRNA in lung tissue of model group was significantly increased on 25,35,45 days of age(P<0.01);Compared with the model group,the relative expression of VEGF mRNA in lung tissue of high-dose and middle-dose of CTCM group and L-Arg group was significantly decreased on 25,35,45 days of age(P<0.01, P<0.05).Compared with the control group, the expression of VEGF protein in lung tissue of model group was significantly increased(P<0.01);Compared with the model group, the expression of VEGF protein in of high-dose of CTCM group and L-Arg group were significantly decreased(P<0.01),the expression of VEGF protein in of middle-dose of CTCM group were significantly decreased on 15,35,45 days of age(P< 0.01).5.Compared with the control group, the relative expression of VEGFR2 mRNA in lung tissue of model group was significantly increased(P<0.01);Compared with the model group, the relative expression of VEGFR2 mRNA in lung tissue in high-dose of CTCM group and L-Arg group was significantly decreased on 15,25,45 days of age(P<0.01),the relative expression of VEGFR2 mRNA of middle-dose of CTCM group on 25,35,45 days of age were significantly decreased(P<0.01),the relative expression of VEGFR2 mRNA in low-dose of CTCM group was significantly decreased at 15,45 days of age(P<0.01).Compared with the control group, the expression of VEGFR2 protein in lung tissue of model group was significantly increased(P<0.01);Compared with the model group, the expression of VEGFR2 protein in of high-dose of CTCM group was significantly decreased(P<0.01,P<0.05),the expression of VEGFR2 protein in of middle-dose of CTCM group were significantly decreased on 25,35 days of age(P<0.01),the expression of VEGFR2 protein in of L-Arg group were significantly decreased on 35,45 days of age(P< 0.01).6.Compared with the control group, the relative expression of VCAM1 mRNA in lung tissue of model group was significantly increased(P<0.01);Compared with the model group, the relative expression of VCAM1 mRNA in lung tissue in high-dose of CTCM group was significantly decreased(P<0.01),the relative expression of VCAM1 mRNA of middle-dose of CTCM group on 15,25 days of age were significantly decreased(P<0.01,P<0.05),the relative expression of VCAM1 mRNA in L-Arg group was significantly decreased at 15,25,35 days of age(P<0.01,P<0.05).Compared with the control group, the expression of VCAM1 protein in lung tissue of model group was significantly increased(P<0.01);Compared with the model group, the expression of VCAM1 protein of high-dose of and middle-dose of CTCM group and L-Arg group were significantly decreased(P< 0.01),the expression of VCAM1 protein in of low-dose of CTCM group were significantly decreased on 15,35 days of age(P<0.05).7.Compared with the control group, the relative expression of SPRED1 mRNA in lung tissue of model group was significantly decreased on 25,35,45 days of age(P<0.01);Compared with the model group, the relative expression of SPRED1 mRNA in lung tissue in high-dose of CTCM group was significantly increased on 25,35,45 days of age(P<0.01),the relative expression of SPRED1 mRNA of middle-dose of CTCM group on 45 days of age were significantly increased(P<0.05),the relative expression of SPRED1 mRNA in L-Arg group was significantly increased on 25,45 days of age(P<0.01,P<0.05).Compared with the control group, the expression of SPRED1 protein in lung tissue of model group was significantly decreased on 25,35,45 days of age(P<0.01);Compared with the model group.the expression of SPRED 1 protein in of high-dose of CTCM group and L-Arg group were significantly increased on 35,45 days of age(P<0.01,P<0.05),the expression of SPRED1 protein in of middle-dose and low-dose of CTCM were significantly increased on 45 days of age(P<0.05).8.Compared with the control group, the relative expression of PI3KR2 mRNA in lung tissue of model group was significantly decreased on 25,35,45 days of age(P<0.01);Compared with the model group, the relative expression of PI3KR2 mRNA in lung tissue in high-dose of CTCM group and L-Arg group was significantly increased on 25,35,45 days of age(P<0.01),the relative expression of PI3KR2 mRNA of middle-dose of CTCM group on 35 days of age were significantly increased(P< 0.05),the relative expression of PI3KR2 mRNA in low-dose of CTCM group was significantly increased at 45 days of age(P <0.05).Compared with the control group, the expression of PI3KR2 protein in lung tissue of model group was significantly decreased(P<0.01);Compared with the model group, the expression of PI3KR2 protein of high-dose and middle-dose of CTCM group and L-Arg group were significantly increased(P<0.01,P< 0.05).9.Compared with the control group,the relative expression of miR-126-3p in lung tissue of model group was significantly increased on 35 days of age(P<0.01);Compared with the model group.the relative expression of miR-126-3p in lung tissue of high-dose of CTCM group was significantly decreased on 35 days of age(P< 0.01).Conclusion1. VCAM1, EGFL7, VEGF, VEGFR2 mRNA relative expression and protein expression increased, SPRED1, PI3KR2 mRNA relative expression and protein expression decreased in lung tissue,miR-126-3p relative expression increased.2 the compound traditional Chinese medicine can down-regulate the expression level of miR-126,VCAM1, EGFL7, VEGF and VEGFR2, can up-regulate the expression level of spredl and PI3KR2, which can be used to control the ascites syndrome in broilers.
