| Allium fetisowii Regel is a membership of Allium. In China, it is one of the only a perennial herb distributed in Xinjiang. A fetisowii Regel has grown in the natural state for a long time, the current research on the biological characteristics is still in blank. The studies on Germplasm about how to responsive environments and how to make the species to survive are still needed to be researched. In this article,we will seek to improve seed germination rate and resistance in the face of adversity the way to its introduction and breeding research to provide reference and measures of dormancy release formulation provides a theoretical guidance through the special geographical environment in arid and semi-arid Xinjiang biological characteristics of seed research of A. fetisowii. The main results are as follows:(1)Study on the seed morphological characteristics showed that A fetisowii Regel seeds were obovate,testa black and surface reticulate; Seed moisture content was 3.95%;Seeds by0.1% TTC viability assay, seed vigor of 82%.(2)The study of law of water absorption and electrical conductivity of seeds showed that Water absorption and soaking time showed a positive correlation. The maximum amount of water weight was 52.28 percent respectively.Seed leachate conductivity changes consistent with the law and the conductivity value indicates, so the seed soaking time is not too long,security soaking time is 36 h.(3)Studies on seed germination characteristics show that seed germination rate at15/25 ℃ variable temperature conditions and germination energy were the highest, which were 26.67% and 17.50%. In continuous darkness germination potential and germination highest index were 20.00%, 10.00% and 0.51. A. fetisowii seeds were in need of variable temperature, light neutral seeds.(4)Studies on seed dormancy characteristics show that seed germination is slow and irregular features from seed embryo morphology after cooked limited, whose sleep is not caused by the sleep of the embryo morphology. After stratification seed treatment can break seed dormancy and variable temperature stratification effect in a constant temperature stratification;Highest germination 60% concentrated H2SO4 treated seed 30 min rate of57.78%. NaOH treatment effect was not significant. The highest seed germination rate ofH2O2 treatment only 7.78%. Multi-seed garlic seed germination process ɑ- amylase activity,(ɑ + β)- amylase activity and protease activity was increased at first and then decreased,restricting the activity of key seed germination, which is the further evidence of multiple seed garlic seed is physiological dormancy.(5)Different concentrations of exogenous substances more and more garlic break seed dormancy has played a very significant role,GA3 150 mg · L-1 for 12 h and the germination rate of 91.00%; 6-BA 5mg·L-1 treatment 12 h, the germination rate was 93.33%;SA 0.1 mg·L-1treatment 14 h, the germination rate was 90.83%;Penicillin300 mg·L-1 treatment14 h of seed germination rate was 84.17%;Wherein the 6-BA treatment of seeds the best.TTC binds activity assay results from the mixing of seed dormancy dormant seed coat and embryo dormancy common disorder caused by physical and chemical methods can break dormancy to some extent under controlled conditions.(6)The research on seeds stress tolerance shows that Hydration- dehydration treatment can promote more seeds garlic seed germination:Artificial aging optimum time is 2h; NaCl and PEG stress treatments significantly impeded seed germination, 300 mmol·L-1NaCl,25%PEG seed under conditions does not germinate.After seeds NaCl and PEG Stress at0.1-0.4mmol·L-1 of SNP treatment1-8h, which can effectively recover NaCl and PEG stress and improve seed germination rate after stress. After NaCl and PEG Stress seed0.1-0.2mmol·L-1SA processing1-8h, which can effectively recover NaCl and PEG stress and improve seed germination rate after stress. Al fetisowii Regel is more tolerant and seed garlic seed medium weak. It can be lifted stress manually during seed germination. |
