| ObjectiveCompound Chinese medicinal herbs(CCMH)was studied on production performance,intestinal microflora and intestinal structure of laying hens,Oxidative enzyme capacity of serum,liver,kidney and yolk by antioxidative activity Kit,bacterial structure of jejunum and the m RNA expression in liver and kidneywas determined by quantitative real-time polymerase chain reaction(PCR).AIM to investigate the effect of CCMH on,antioxidant capacity,production performance of laying hens.MethodsThe jejunum of laying hens was made parafin section according to HE staining technique.The villus length,axillary depth,the height of villi and crypt depth were measured with Olympus microscope BX53 and software Image-Pro Plus6.0.The enzyme activity of SOD,GSH-Px and MDA was detected with Regent Kits,and the expression of.GSH-Px and SOD1 m RNA in liver and kidney were determined with real-time PCR technique.Objective to study whether CCMHcan enhance the activity of anti oxidative enzymes to stabilize the structure of the intestinal flora and to improve the production performance of the layer by regulating the expression of the anti oxygen enzyme gene.Results1.CCMH improved laying rate,average egg weight,and feed ratio of laying hens.2.The number of caecal lactobacilli and bifidobacteria in laying hens increased,and the number of cecal Escherichia coli,fecal coliform bacteria decreased(p<0.05).The jejunal villus height,crypt depth,the height of villi and crypt depth(V/C),and mucosal thickness decreased also(p<0.05)3.The enzyme capacities of SOD and GSH-Px in serum and kidney were improved(p<0.05),MDA level in serum,yolk,and kidney decreased(p<0.05).4.The results shows that the expression level of SOD1 and GSH-Px in the kidney is significantly affected by CCMH.Compared with the control group,m RNA expression level of GSH-Px was significantly increased in the group of 50g/t CCMB,as well as the m RNA SOD expression in 100mg/t group(P<0.05),150mg/t up the m RNA GSH-Px expression.Conclusions1.It is conclued that supplementation of CCMH to laying hens helped to maintain the stability of intestinal flora.The number of caecal lactobacilli and bifidobacteria in laying hens increased,and the number of cecal Escherichia coli,fecal coliform bacteria decreased.2.The jejunum villus length,crypt depth,intestinal muscle thickness and V/C values were significantly improved.It indicated that CCMH can help villous atrophy caused by oxidative damage improve intestinal digestion and absorption of nutrients,and improve the performance of laying hens.3.CCMH improves the activity of antioxidant enzymes in serum,kidney,and reduce the malondialdehyde(MDA)content in egg serum,egg yolk,and kidney.The activities of SOD1 and GSH-Px in liver and kidney changed with its m RNA expression level.It is concluded that antioxidative capacity closely related to antioxidantgenes expression. |
