Isolation, Identifiation And Biological Characteristic Research Of Streptococcus From Pig In Anhui

Streptococcus suis diseases is an infectious diseases caused by all kinds of Streptococcus,there are significant differences between different types of pathogenic characteristics.The advantage strain is a dynamic process in different area or time,the characteristics of this complex pathogen brings difficulties to its prevention.In this study,PCR method was used to identify the swine Streptococcus which isolated in Anhui during 2009 ~ 2014 year,1,2,7,9 type and ef,mrp,sly virulence genes;VITEK 2 advanced automatic identification equipment was used to identify non-Streptococcus suis;PFGE technique was used to identify genetic typing of the advantage bacteria;K-B disc diffusion method was used to detect the drug sensitivity.Aims to understand the swine Streptococcus biologyical characteristics in Anhui,to build preliminary PFGE database source of swine Streptococcus and discusses the connection between the swine Streptococcus in Anhui,To provide scientific basis for tracing to the source of swine Streptococcus disease pathogens and studies its molecular epidemiological in Anhui,it is of great significance for swine Streptococcus disease prevention and control in Anhui.Results: Among 196 strains swine Streptococcus,112 strains Streptococcus suis(57.1%)belongs to the dominant bacteria,the rest 40 strains belongs to Enterococcus,accounting for 20.4%.Of the 112 strains Streptococcus suis: 44 strains belongs to type 2,accounting for 39.3%,which was the superiority serotype;ef,mrp,sly virulence genes carry rate were 39.3%,36.6% and 39.3% respectively,ef-/mrp-/sly-(36 strains,32.1%),was the advantage virulence genotype,Streptococcus suis type 2 virulence genes carry rate of ef,mrp,sly is significantly higher than that of Streptococcus suis,and were 75.0%,52.3%,88.6% respectively,ef+/mrp+/sly+(19strains,43.2%)was advantage virulence genotype;Streptococcus suis consist of 56 PFGE genotype,Ps28(18 strains,16.1%)and Ps27(14 strains,12.5%)are advantage PFGE genotype,and the two PFGE genotypes mainly are Streptococcus suis type 2;Only to vancomycin,ampicillin,amoxicillin,levofloxacin,and cephalosporins sensitive rate were above 50.0%,to Tetracycline,sulfamethoxazole,streptomycin,doxycycline,gentamycin,azithromycin,and cotrimoxazole resistant rate were above 85.0%,the resistance tendency is distinct to penicillin,ceftriaxone,cefotaxime,cefepime,multiple drug resistance is serious,more than 8 kinds of drug resistant 96 strains,accounting for 85.7%.Among 40 strains of Enterococcus,22 strains Enterococcus faecium,accounting for 55.0%,which was divided into 20 PFGE genotype,Pe12(9 strains,22.5%)and Pe7(7strains,17.5%)were advantage PFGE genotype,and the two PFGE genotypes mainly were Enterococcus faecium.Only to vancomycin,teicoplanin sensitive rate above 72.0%,Streptomycin,trimesulf,sulfamethoxazole,gentamycin,cefepime,tetracycline,azithromycin and ceftriaxone resistant rate of 85.0%,multiple drug resistance is serious,more than 8 kinds of drug resistant 38 strains,accounting for 95.0%.Conclusion: Swine Streptococcus in Anhui area was really complex,Streptococcus suis was the dominant bacteria and type 2 was the superiority serotype,ef-/mrp-/sly-was the advantage virulence genotype,Enterococcus have become the important pathogens in swine industry in Anhui area.Streptococcus suis and Enterococcus PFGE genotypes were polymorphic.The PFGE genotypes with serotype,virulence genes don’t present correlation,Streptococcus suis type 2,Enterococcus faecium were mainly from two clones,and propagation exist genetic variation.Streptococcus suis and Enterococcus drug resistance,multi drug resistant spectrum is becoming more and more wide.
