| In this study, we observed the effect eight Chinese medicines addition when culturing Sheep Ovarian particles cell in vitro. Using molecular biology, particularly real-time PCR, we tested the expression changes of apoptosis genes such as Bax, Bcl-2 after chlorpromazine addition, H2O2 exposure sheep ovarian granulosa cells. We studied the promotion effect of eight tonic medicine on sheep ovarian granulosa cell proliferation and the protective effect of sheep granulosa cells after exposure to the.The results are as follows:1. The most appropriate density of cultured granulosa cells was about 106/ml, the cell growth turned to slow down when below 103/ ml, and began to change in morphology.2. Astragalus, Acanthopanax, longan, wolfberry, Ligustrum lucidum, Gecko and dodder promoted granulosa cells proliferation, but foxglove had no significant effect in proliferation promotion.3. Chlorpromazine significantly inhibited granulosa cells proliferation when above 0.1μmol / L in concentration. H2O2 significantly inhibited granulosa cells proliferation when above 150μmol / L.The Bax, Bcl-2 expressions were up regulated together with increasing of the chlorpromazine and H2O2 concentrations.4. Astragalus, Acanthopanax, foxglove, longan, wolfberry, Ligustrum lucidum, Gecko, dodder inhibited the cell-damaging effect of chlorpromazine. Astragalus, Acanthopanax, foxglove, longan, wolfberry fruit, Ligustrum lucidum, Gecko, dodder inhibited the cell-damaging effect of chlorpromazine of H2O2. |
