| Pork is the source of animal protein of the Chinese consumer who followed with interest for the meat quality. The fat content in pork is an important factor for the formation of quality and flavor, determining the juiciness and taste of pork. Genetic factors regulating the fat deposition in pork is closely related to THRSP gene. The levels of the gene expression related to fat synthesis and decomposition are associated with the polymorphism of THRSP gene.Dingyuan pigs were employed as experimental animals.5 SNP sites were detected in the promoter region of THRSP gene. The recombinant plasmids with 5 SNP combined-types were constructed for double luciferase reporter gene, and then co-transfected with pRL-CMV plasmids to 293T cells. The different promoter efficiencies were tested using Luciferase Assay System, and analyzed in the association with the expression level of the THRSP gene.The results showed that the 5 SNP sites located in A-95T, G-98A, C-376A, T-400C and C-459T upstream from the CDS start of the THRSP gene, the promoter region of THRSP gene.the TCAGA promoter efficiency from 5 SNP loci was highest. The efficiency of TCAGA promoter increased 129% than that of CTGGA. Compared with CTGGA, the promoter efficiency of CCAGA, CTAGA and CTGAT were lower 99%,73.2% and 42.6%, respectively. The promoter efficiency of TCAGA was 75.6% higher than that of CCAGA, and CCAGA was 96.6% higher than that of CTAGA. The promoter efficiency of SNP was revealed by reporter gene as the results by real-time PCR gene for THRSP gene expression.The expression level of the THRSP gene was significantly correlated to the promoter efficiency (P<0.05), and are consistent with those of the allele frequency distribution in lean pigs. The results implied that the polymorphism at A-95T, G-98A, A-376G, T-400C and C-459T were the key factors for the differences among the expression levels of THRSP gene. |
