In this paper,Angelica sinensis,Radix Astragali,Codonopsis pilosula,and licorice are used as raw materials,and various enzymes are added to ferment and prepare functional enzymes of Chinese herbal medicine.The optimum fermentation technology is determined by protease activity,total flavonoid content and total sugar content,and the compound process is studied by adding red ginseng,oleuropein and propolis.Using electrochemical method to study the scavenging ability of different concentrations of functional enzymes to hydroxyl radical,superoxide free radicals,lipid peroxide free radical,DPPH free radical.The types and contents of some effective active substances in functional enzyme are quantitatively analyzed by high performance liquid chromatography.The results are as follows:First,the ratio of 1:3:1 cellulase,amylase and glucoamylase are weighed and ferment for 48 h at 37°C and 60 r/min shaking speed as the optimu m conditions for the fermentation.The protease activity is 1.7288 u/m L and total flavonoids are obtained 0.1140 mg/m L,total sugar 2.7833 mg/m L.The protease activity of the functional enzyme after compounding is 2.3810 u/m L,total flavonoids 0.2069 mg/m L,and total sugar 3.6200 mg/m L.The protease activity of the functional enzyme after compounding is increased by 0.6250 u/m L over that of the functional enzyme before compounding,the total flavonoid content is increased by two times,and the total sugar content is increased by 0.8400 mg/m L.Second,using cyclic voltammetry to study the scavenging ability of different concentrations of functional enzymes to hydroxyl radical,superoxide free radicals,lipid peroxide free radical,DPPH free radical,and calcu late clearance rate respectively.The functional enzyme complexes improve the clearance rate of four kinds of free radicals,and reduce the minimum clearance concentration of four kinds of free radicals.The scavenging effect of functional enzyme complex is the best for the scavenging effect of superoxide free radical and lipid peroxide free radical.DPPH and hydroxyl radical are followed.Third,the types and contents of ferulic acid,z-ligustilide,astragaloside A,lobetyolin,liquiritin,oleuropein and Ginsenoside Re in functional enzyme are quantitatively analyzed by high performance liquid chromatography.The content of active substance in functional enzyme: liquiritin > z-Ligustilide > ferulic acid > lobetyolin > astragaloside A > oleuropein > ginsenoside Re.
