Hericium erinaceus polysaccharideshavethe functions of anti-cancer,healing of gastritis and gastric ulcer,lowing blood sugar and other biological activity.In order to improved the technology for extracting Hericium erinaceus polysaccharides,this paper aimed to study ultrasonic-assisted and multi-stage dynamic countercurrent extraction and processing engineeringfor extracting polysaccharides from Hericium erinaceus fruit body to increase the yield rate.The new extraction processing was applied on an industrial scale for production of Hericium erinaceus oral liquid.In addition,a rapid and accurate chromatographic method was established for analyzing molecular weight of extracted polysaccharide.The main contents and results were as follows:1.The conditions of the ultrasonic-assisted and multi-stage dynamic countercurrent extractionprccessing was optimized based on the single tank extraction for Hericium erinaceus polysaccharide with orthogonal experiments.The results showed that:An extraction process assembled by three tanks in continuous dynamic countercurrent mode and Hericium erinaceus treated at 1:15 of solid-liquid ratio,and 50?for 2 hours was established,and the extraction was enhanced by ultrasonic at 400 W for 25 minutes.The results showed that polysaccharide extraction rate reached about 13.37%.Compared to the traditional method of single tank extraction,the extraction rate of Hericium erinaceus polysaccharide was increased by more than 50 %,and extraction time and used solvent were decreased by about 30% and 40% respectively.2.High performance gel filtration chromatography(GFC)and differential refraction detection were used to performe the molecular weight and quantitive analysis on the extracted polysaccharide from the Hericium erinaceus fruit bady and Hericium erinaceusas oral liquid.based onthe retention time and the peak area external standard method The results showed that polysaccharide of Hericium erinaceus and Hericium erinaceus oral liquid had the molecular weights from 16,000 Da to 17,000 Da,and wide D distribution from 1.05 to 1.07.Polysaccharide content of the average was 0.66% in Hericium erinaceus and 37.1mg/mL in the oral liquid.After 75% ethanol precipitation,gel chromatography proved that using 33%~50% ethanol to pretreat crude extract of Hericium erinaceus polysaccharideAnd combined with alkaline copper reagent could effectively purify product of Hericium erinaceus polysaccharide.3.One production linewith output of 1,000 tons Hericium erinaceus oral liquid annually was designed with the technology of ultrasonic-assisted and multi-stage dynamic countercurrent extraction based on the engineering,The main contents including process design,material balance,equipment selection,establishment of quality standards,investment estimates,profit margins estimate and so on.A new production workshop was designed for extracting polysaccharide from Hericium erinaceus fruit bodybased on theprocessing of ultrasonic-assisted and Multi-stage dynamic countercurrent.Compared to the existing traditional production process,the new technology shows the merits in saving dried Hericium erinaceus by 54.6% to produce the same quantity of Hericium erinaceus oral liquid in addition,the ales profit rateswascalculated comparatively higher by 50.8% and the overall profit of Hericium erinaceus oral liquid will be by 15% higher than the traditional processing based on the perspective of financial analysis.
