In this paper,a new type of experiment for preparation of hollow fiber membrane chro-matography was designed.In order to obtain hollow fiber membrane with suitable structure and large specific surface area,the effect of coagulations on the membrane microstructure was investigated,and the dimension of hollow fiber membrane was regulated by changing the dry-wet spinning operation conditions.In the end.the hollow fiber membrane chromatography was prepared,and used for the adsorption of the protein.The researches and results are as follows:First of all,using N-Methyl pyrrolidone(NMP)as solvent,CTA flat membranes were pre-pared with different first coagulations:water/NMP,ethylene glycol(EG)/NMP,ethanol/NMP and glycerol(Gly)/NMP,and water as second coagulation.The membranes were character-ized and then prepared into chelated copper ion membrane chromatography(MC),respectively.Membrane structure and adsorption properties of copper ions and proteins of membrane chro-matography were determined.The SEM pictures revealed that membranes prepared in Water/NMP and EG/NMP has dense surface and large voids in the cross-section.Gly/NMP membrane has spongy-like pores which connected into a network structure.Surface roughness changes in the order of MC-Gly>MC-EtOH>MC-EG>MC-Water.Besides,MC-Gly membrane chro-matography adsorped the highest amount of copper ions,and MC-EtOH membrane chromatog-raphy adsorbed the highest amount of amount of lysozyme and BSA.On the contrary.MC-Water membrane chromatography adsorbed the lowest amount of lysozyme and BSA.Secondely,the CTA hollow fiber membrane was prepared by dry-wet spinning method,and the effect of spinning parameters on the structure of hollow fiber membrane was explored.The results show that inner and outer diameter of hollow fiber membranes increased with the increase of dope and bore flow rate,and decrease with the increase of take-up speed;the inner diameter decreased with the increase of air gap distance.Besides,pure water flux of hollow fiber membranes increased obviously with the increase of dope and bore flow rate,and increased slightly with the increase of take-up speed and air gap distance.The minimum inner diameter of the CTA hollow fiber in this study is about 20 ?m.Finally,a new kind of hollow fiber membrane chromatography was designed,and the ad-sorption of copper ions and BSA were tested.
