There are abundant biological resources in the ocean and certain particularity of marine environment result in marine organisms polysaccharides with novel structure and some other special characteristics.In order to enrich the physiological activity of marine polysaccharides,the study was conducted on the Polysaccharidic Extract from Sea Cucumber Processing Liquor(PESCPL),Polysaccharidic Extract from Scallop Skirt(PESS)and Polysaccharide of Sporopyll of Undaria Pinnatifida(PSUP).On the one hand,the PESCPL was used in the RAW264.7 cell system to detect its immunoregulatory activity,antioxidant activity and antiinflammatory activity in vitro,thus reflecting its contribution to the immune system.In order to detect the above physiological activity,some indexes including the phagocytosis ability,NO release ability,proinflammatory cytokine secretion rate,ROS production and antioxidant enzyme system damage were analyzed in the normal cell system,inflammatory cell system and oxidative damage cell system separately treated with different concentrations of PESCPL.On the one hand,the polysaccharide was extracted from the Sporopyll of Undaria Pinnatifida and optimized its extraction process.At the same time,the polysaccharide was used in the RAW264.7 cell system to detect its antioxidant activity.On the other hand,the PESS was extracted and the main components were determined.In addition,the extracted PESS was also analysed in RAW264.7 cell system to detect the antioxidant activity.By analyzing the three aspects above,the possible contribution of marine polysaccharides to the immune system could be reflected.The results showed that the PESCPL could regulate the immune response of RAW264.7 cells by regulating the NO release ability,PGE2 release ability,phagocytosis and iNOS activity.Compared with the control,the NO release capacity,PGE2 release ability,phagocytosis and iNOS enzyme activity were increased by 21.8,6.7,1.2 and 3.2 times,respectively,by the maximum concentration of PESCPL.Secondly,it had good antioxidant effect,including the mitigation of cell oxidative damage caused by H2O2,the viable cell survival rate was 1.7 times higher than that of oxidative damage.In addition,it also had good influence on anti-inflammatory,including the levels of TNF-?,IL-1? and IL-6 in the LPS-treated cells,which were reduced to 0.2,0.2 and 0.7 times of the LPS alone treated group.Compared with the LPS alone group,the release capacity of NO was inhibited by approximately half,the enzyme activity of iNOS was reduced by 0.83 times,and the expression of COX-2 was decreased.The research provides some theoretical basis and reference direction for the future development of comprehensive improvement of immunity function food,and has certain potential value.The pretreatment conditions of papain-assisted extraction of PSUP were as follows:the mesh size was 120 mesh,the immersion time was 12 hours,and the soaking temperature was 50 ?.The optimum conditions were as follows:enzymatic hydrolysis time was 6 h,enzyme dosage was 2.45%,liquid ratio was 79:1,and the rate of PSUP was 7.96 ± 0.86%.The PSUP has good antioxidant effect,which can alleviate the oxidative damage caused by H2O2,decrease the content of ROS and MDA in oxidative damage cells,and increase the enzyme activity of SOD,CAT and GSH-Px in oxidative damage cells.The research provides the theoretical basis for the standardization production of the enterprise,and provides some theoretical basis and reference direction for the future development of the antioxidant functional food which has certain potential value.The main active ingredient of PESS was carbohydrate.It had good antioxidant activity,including that it decreased the content of MDA and ROS,increased SOD,CAT and enzyme activity of GSH-Px.This study has proven that these three marine polysaccharides all had good immunoregulatory activity,which can be used as accessories or principal material.Moreover,it provided a new road for the development of potential value.
