Chlorogenic acid(CGA)and caffeine are two important bioactive substances in coffee,our previous studies found that CGA+caffeine can significantly reduce the weight and IPAT of mice.The aim of this experiment is to further explore the influence of CGA and caffeine combination on the lipid metabolism in high fat-diet(HFD)induced obese mice.80 female ICR mice were equally divided into 8 groups:control group(I),HFD(II),HFD + 0.2% CGA(III)?HFD + 0.04% caffeine(IV)?HFD + 0.1% CGA + 0.02% caffeine(V)?HFD + 0.1% CGA + 0.04% caffeine(VI)?HFD + 0.2% CGA + 0.02% caffeine(VII)?HFD + 0.2% CGA + 0.04% caffeine(VIII),fed for 14 weeks respectively.Body weight and food intake were determined during this period.Organs and intraperitineal adipose tissues(IPAT)were weighted after the feeding period.Serum biochemical indexes and liver lipids contents were measured.Liver and IPAT were observed pathologically with hematoxylin and eosin(HE)staining and the routine paraffin techniques.The hepatic m RNA and protein expression of lipid metabolism related enzymes were determined.Results were as followed:(1)The weight gain of IV,VI-VIII group and IPAT weight of IV,VI,VIII group were significantly reduced compared with II group.(2)Compared with II group,the serum TG,LDL-C and liver TG levels of VI-VIII group were significantly reduced,TC levels in serum and liver of IV,VI-VIII group were significantly reduced,serum FFA was significantly reduced in VIII group.(3)Compared with II group,serum leptin were reduced significantly,while adiponectin were significantly increased in VI-VIII group;serum interleukin-6significantly was depressd in VIII group mice.(4)Observed by the HE staining and the routine paraffin,compared with II group,fat droplets in liver of VI-VIII group were more,and the diameter of fat cells in IPAT mice of IV,VI and VIII group were smaller.(5)The CGA and caffeine combinations inhibited the mRNA expression of HMGR,SREBP1 c,FAS,SCD1 and LXR? in the liver,while promoted the m RNA expression of AMPK?,HSL,ATGL,ACO,Apo-E in liver.(6)The CGA and caffeine combination inhibited the protein expression of FAS,SCD1,LXR?,and promoted the protein expression of AMPK?,HSL,ATGL,and Apo-E in liver.The results showed that CGA and caffeine combination regulated the mRNA and protein expression of lipid metabolism related genes in mice liver,then inhibited fat synthesis,promoted lipid oxidation,so as to achieve the slimming effect.The combination effects were better than CGA or caffeine alone,therefore CGA and caffeine have synergistic effect on lipid-lowering.
