Bacteriocin is biodegradable.It is considered a safe and natural biological preservatives.There are four kinds of food preservative films in this study,polyethylene(PE),high density polyethylene(HDPE),low density polyethylene(LDPE)and polyvinylidene chloride(PVDC)films,respectively.To preparation of bacteriocin biological preservative films and applied of cold meat to control the microbial contamination,the specific research contents and results are as follows:(1)Preparation of biological preservative filma.The films were cutted into circles with diameters of 10 mm and soaked in 10 mL of plantaricin BM-1 solution(20480 AU/mL)for 2 hours.After soaking,the films were removed from the solution.The antibacterial activities against L.monocytogenes on TSB-YE plates of biological preservative films exhibited obvious inhibition zones around the film.The results showed that the diameters of the inhibition zones around the PVDC-BM-1 films most obvious were 16.2 mm.b.Four kinds of films were immersed in Lactobacillus plantarum BM-1 solution respectively,and the adsorption amount of plantarum BM-1 was observed with different immersion time.The results showed that the adsorption capacities were about 420?L-430?L,And there was no significant difference between the maximal adsorbents(p <0.05).c.The four kinds of biological preservative films were soaked in sterile water to analyze the relationship between the desorption capacity of Lactobacillus plantarum BM-1 from the film with the soaking time.The results showed that PE-BM-1,LDPE-BM-1 and PVDC-BM-1 were immersed in sterile water for 16 h,20 h and 24 h respectively,and the maximum desorption capacity was 2560 AU/mL,while HDPE-BM-1 in sterile water 12 h to achieve the maximum desorption capacity,the maximum desorption capacity of 1280AU/mL.d.The four kinds of films were immersed in Lactobacillus plantarum BM-1 solution at different times to make biological preservative films.After storage for a period of time,the antibacterial activity was tested and the stability of the biological preservative film was analyzed.The results showed that the four kinds of films were immersed in the solution of bacteriocin BM-1 for different time,and stored at 25 ? for different days.Listeria monocytogenes were significantly inhibited.Indicating that the biological preservative films have a high stability at 25 ?.(2)Application of PVDC-BM-1 in microbial contamination control of the chilled meata.Artificial Listeria monocytogenes of 3.34 lg CFU/mL and 4.20 lg CFU/mL on the meat's surface.Over the 7-day cold storage period,the counts of L.monocytogenes in the PVDC-BM-1 active packaging group increased from an initial 3.34 lg CFU/mL to a maximum count of 3.71 lg CFU/mL.When the initial inoculation of L.monocytogenes was 4.20 lg CFU/mL in chilled meat.The counts of L.monocytogenes in the PVDC-BM-1 packaging group increased from an initial 4.20 lg CFU/ml to a maximum count of 4.52 lg CFU/ml over the 7-day cold storage period.All of these results suggested that the plantaricin BM-1-activated PVDC film could significantly inhibit the growth of L.monocytogenes in chilled meat during cold storage.b.The total bacterial counts of the chilled meat were 3.64 lg CFU/g,which was gradually increased to 4.26 lg CFU/g.The pH and TVB-N values were increased to 5.82 and 11.2 mg/100 g,respectively,after 7 days of storage at 4?.In meat treated with PVDC-BM-1,pH and TVB-N contents were significantly lower(p<0.05)than that of the PVDC film groups at the same time points during storage,which indicated that the PVDC-BM-1 films extended the shelf life of the meat samples during cold storage.
