Jimai-22 has some excellent characteristics with high yield, good disease resistance and widet adaptability. It has become the most popular wheat variety in China since 2010. Presently, wheat bran, the main by-product, is mainly uesd for animal feed with lower economic value. In this study, feruloyl xylo-oligosaccharides in the bran from Jimai-22 was extracted and its physiological activity was preliminarily researched. I hope this study would provide a reference for the application of feruloyl xylo-oligosaccharides in functional foods, medicine and cosmetic, and improve the research and development of the bran from Jimai-22.1.The antioxidant capacity of enzymatic hydrolysate from 6 different species of bran was determined. It was found that the enzymatic hydrolysate of the bran from Jimai-22 has the lowest IC50 values of ABTS and DPPH radical scavenging capacities among these species. Therefore, Jimai-22 was selected for further study.2.The process of extracting feruloyl xylo-oligosaccharides of water unextracted polysaccharides from Jimai-22 by the xylanase was studied. The optimum process was determined as temperature 43.78 ?, pH 5.1, time 44.62 h, enzyme concentration 4.86 g/L and substrate concentration 87 g/L. In this process, the concentration of feruloyl xylo-oligosaccharides was 1.326 ± 0.020 mmol/L.3.The antioxidant capacity of feruloyl xylo-oligosaccharides in vitro was determined by DPPH, ABTS, FRAP and hydroxyl radicals scavenging reducing experiments. The results showed that the antioxidant activity of feruloyl-xylo-oligosaccharides was potent and was stronger than ferulic acid.4.The inhibitory effect of feruloyl xylo-oligosaccharides on common bacteria in food, such as S. aureus, B. subtilis, E. coli, S. typhi, S. cerevisiae and M. luteus, was determined by their inhibition zone diameter and minimum inhibitory capacity. The results showed that feruloyl xylo-oligosaccharides had certain inhibitory effect on the above several bacteria but had no effect on S. cerevisiae and M. luteus.5.The prebiotic effect of feruloyl xylo-oligosaccharides was determined by mesuring their proliferative effect on lactobacillus etc.The result showed that feruloyl xylo-oligosaccharides had certain prebiotic effect on Lactobacillus but their prebiotic effect on probiotics was slightly weaker than Xylo-oligosaccharides.6.The enzymatic hydrolysate was purified successively by Amberlite XAD-2 resin and Sephdadex LH-20 resin. Then the purified sample was analyzed and identification by HPLC and HPLC-MS, and 3 compounds were idetified respectively as Xyl-(1,4)-(F-Ara-(1,3)-Xyl-(1,4)-Xyl-(1,4)-Xyl), Xyl-(1,4)-(F-Ara-(1,3)xyl-(1,4)-Xyl) and ferulic acid combined with the relevant literature.
