As a kind of plants' secondary metabolites,flavonoids colour fruits and flowers,favouring seed and pollen dispersal,and contribute to plant adaptation to environmental conditions and pathogen attacks.At the same time,they could help for body heaithy.There are many enzymes in the phenylpropanoid pathway that catalyze the substrate into many derivatives,for example chalcone isomerase(CHI).A key branch-point enzyme of the phenylpropanoid pathway,CHI,catalyzes the reaction producing flavanones,the skeletal backbone for many downstream metabolites.This reaction is conducted by the cyclization of naringenin chalcone to naringenin.Here,we identified an additional genes in the CHI family from Caragana intermedia,The results are as follows.1.The full length gDNA sequence of CiCHIL gene was 1434bp,cDNA sequence of CiCHIL gene was 941bp,consist of ORF(630bp),5'-UTR(164bp)and 3'-UTR(147bp).The deduced protein comprised 209 amino acids,which contains a 12 adenylate composition of the Poly A tail,a total of 4 exons and 3 introns.2.Binary vectors p35S::CiCHIL were constructed and the homozygous transgenic Arabidopsis thaliana overexpressing CiCHIL were obtained.3.The content of total flavonoids were detected.We found that the content of total flavonoids in Arabidopsis thaliana overexpressing CiCHIL was higher than wild type?4.The expression of CiCHL under UV-B stress was detected by qRT-PCR,CiCHIL was induced by UV-B.After UV-B stress,overexpressing lines could still maintain higher resistance.
