Genes of UF2 encoding a protein contained conserved domain of DUF(domain of unknown function) 1644 responses to sugar signal in the process of seed germination. RGS1(regulator of G-protein signalling), a sugar sensor, plays an important role in the perception of sugar signal and the signal transduction. RGS1 is composed of seven transmembrane domains and a C-terminal box in Arabidopsis. Bioinformatics indicates that UF2 might interact with AtRGS1. In this study, we investigated the relationship between UF2 and AtRGS1. The results are as follows:(1) Bimolecular fluorescence complementation(BiFC) experiment showed that UF2 can interact with AtRGS1;(2) The GAL4 system yeast two-hybrid(Y2H) experiment, BiFC results and Pull down results showed that UF2 interacted with AtRGS1-box.(3) Meanwhile,the DUF1644 of UF2 did not interact with AtRGS1-box in our Y2 H experiment.We also investigated another gene encoding a protein of unknown function, UF1,contained conserved domain of DUF584. BiFC experiment showed that UF1 can inteact with AtRGS1; Y2 H and BiFC experiments showed that UF1 interacted with AtRGS1-box. But the result of Y2 H experiment showed that DUF584 of UF1 did not interact with AtRGS1-box.Tetraspanin(TET) family proteins have significant functions during the process of growth and development in animal. But we know little about the functions of TET in plant species although they are widely distributed. Arabidopsis TET6(AtTET6) responses to sugar signal in the process of seed germination in our previous results. In this research, we investigated the subcellular localization of AtTET6. The transient expression of35S::AtTET6-GFP in tobacco leaves and stable expression of AtTET6pro::AtTET6-GFP in Arabidopsis tet6 showed that AtTET6 was located in the endoplasmic reticulum(ER).
