In recent years, with the development of fabricating technologies of microfluidic chip, fabricating technologies of nanofluidic chip or micro-nanofluidic chip has also been rapidly developed. Micro-nanofluidic chip is used in the research such as ion or molecular analysis, separation and enrichment, so it has broad prospects. With an increasing smaller size of nanochannel flow, nanofluidic chip has some special properties such as surface electrical charge, the electrical double layer, increased viscosity and decreased electroosmotic flow. These special properties make micro-nanofluidic chip play an important role in biology and chemical analysis. In these important applications, the working electrode plays a very important role.In this paper, according to the features of the microfluidic chips and nanofluidic chips, the structure of the micro-nanofluidic chip control experimental device for the bio-molecule manipulation is designed. The nanofluidic diodes and triodes are the structure units of the micro-nanofluidic chip. And in accordance with the functions of the nanofluidic diodes and triodes, the structures of their electrodes are designed, respectively. The control electrodes are fabricated with right method, and they are detected, too. It is very useful for the design and fabrication of the electrode of the bio-molecule manipulation chip.Then, the functions, structure and size parameters of bio-molecule manipulation chip are designed, and according to the structure of the chip, the structure, parameters and functions of the electrode are described. The three-dimension electrostatic field of the electrode is simulated by ANSOFT. Because of the electrode lead, the electric field strength in the nanochannel is "parabolic" shape. After improving the structure of electrode, the electric field strength of different structures electrodes are compared, and the best structure of electrode is found. The movement of bio-molecules in the nanochannel is simulated by GROMACS. It is useful for the manipulation experiment of the bio-molecules in the future.The materials and fabrication method of the control electrode of bio-molecules are chosen. The control electrode of bio-molecular is fabricated by DC magnetron sputtering. The electrode is also detected, and the electrode meets the design requirements.Finally, according to the experimental requirements, the micro-nanofluidic chip control experimental device of bio-molecules manipulation is set up with the micro-nanofluidic chip. The DNA molecules solution is injected into the chip, and the movement of the DNA molecules is observed under pressure with the microscope. The DNA molecules are detected in the chip. After discussing and analyzing the results of the experiment, the experimental device is feasible.
