Flos Puerariae, the flowers of Pueraria lobata (Willd.) Ohwi and P. thomsonii Benth., is an important herbal medicine used to ameliorate ethanol intoxication in Chinese Traditional Medicine. In this thesis, the quality standard of different species and materials collected from different habitat were studied. Furthermore, the flavonoid constituents in the P. thomsonii Benth. were studied, and preparetion of kakkalide as the raw medicine was studied. The following studies were carried out.1. The contrast studies were carried out for the first time at characters, microscopic, physico-chemical aspects.2. The qualitative identification between Pueraria lobata (Willd.) Ohwi and P. thomsonii Benth. was developed at morphology and microscopic identification aspects, and the detail pictures were showed by digital photography for the first time.3. Rapid and clear TLC and HPLC analysis of the flowers of P. lobata and P. thomsonii were developed using kakkalide, tectorodin and tectorigenin as marker compounds. There were significant differences in the contents and main components of isoflavones between the flowers of P. lobata and P. thomsonii.7 different species and collected from different habitat were determined, the result showed out that kakkalide is the main flavonoid constituts of P. lobata while tectoridin and tectorigenin are the main compounds of P. thomsonii.4. The flavonoid constitutens in P. thomsonii Benth. was studied for the first time,11 compounds were isolated, among which 9 were isoflavone,2 was flavone, whose structures were determined as 4’,7-dimethyltectorigenin (1), iristectorigenin A (2), irisolidone (3), genistein (4), irilin D (5),7-methyltectorigenin (6), tectorigenin (7), tectoridin (8), kakkalide (9), luteolin (10), apigenin (11). Compounds 2,5 and 6 were isolated from Pueraria genus for the first time, and compounds 1,3,4 and 7~11 were isolated from the flowers of Pueraria thomsonii Benth. for the first time.5. The extraction technology of kakkalide was developed by orthogonal experimental design, combined with purification technology such as recrystallization, the preparation technology of kakkalide was establishmented. The puerity of kakkalide prepared reached above 95%. By physical,chemical and spectral analysis, the structure was identified. Raw medicine quality standard of kakkalide was developed.
