Studies On Anti-inflammatory And Analgesic Pharmacodynamics Of Zonk 108 Injection And Its Mechanism

The major component of Zonk108 injection is a single chemical composition, and it belongs to non-steroidal anti-inflammatory drugs. Zonk108 was screened in this study by xylene-induced mice ear swelling model and writhing-induced mice by acetic acid model, and exert a good anti-inflammatory and a good analgesic effect. After that multiple kinds of animal models were used to evaluate its anti-inflammatory, analgesic, relieving fever effects, toxicity and mechanisms.The study on anti-inflammatory effect shows that, Zonk108 (iv, the same below) has potent inhibitory effect on xylene-induced ear swelling in mice. Zonk108 has potent inhibitory effects on vascular permeability increase caused by acetic acid and carrageenin-induced hind paw edema. But it has no effect on the primary effect of adjuvant arthritis.The study on analgesic effect shows that, Zonk108 has potent inhibitory effect on writhing-induced by acetic acid in mice, and its effects are similar with Flurbiprofen. has potent inhibitory effect on xylene-induced ear swelling in mice Zonk108 also possessed potent analgesic effect and showed effective anti-nociception in the amount of the time spent in licking the injected paw was significantly reduced in the formalin test in the late phase. 192mg/kg of Zonk108 could also prolong the elapsed time of mice on hot-plate hyperalgesia assay and demonstrated central analgesic effect. And Zonk108 could increase the pain threshold of carrageenin- -induced bind paw edema by pressure in rats. But it has almost no effect of heat stimulus in mice or the electrical stimulation in mice.The study on relieving fever effect shows that, 2.8-25.2mg/kg of Zonk108 has significant effect on febrile body temperature in rats.The study on mechanism shows that 75.6mg/kg of Zonk108 could decrease the amount of PGE2 and NO in exudation of rats, but it had no significant effect on MDA, SOD, 5-HT, His. Zonk108 could decrease the amount of vitamin C and stimulate the function of cortex glandulae suprarenalis with effect like ACTH. The results of safety experiment show that, Zonk108 hasn’t hemolysis reaction to red blood cells from rabbit, and the muscular tissue stimulus and the blood vessel stimulus in rabbit were all negative. The test of initiative hypersensitive reaction suggested that Zonk108 had no allergization in cavia cobayas.In conclusion, Zonk108, a new drug with excellent anti-inflammatory, analgesic and relieving fever effects, which has lower toxicity and exploitation value.
