| Woridwide, lung cancer is not only the most common tumor, but also the tumor of supereme incidence and death rate in all malignant tumor. Recently, operation and chemotherapy are reported as the main means of malignant tumor treatment. But clinical chemotherapy drugs not only damage the tumor cells but also impair the normal cells. Therefore, the treatment can’t be carried out successfully. In this study, Spica Prunellae was selected as a model medicine. Structural Units with chemoprevention and anti-lung cancer activity was screened for the first time under guidance of Material Basis Components Structure Theory in vitro and in vivo, and with its anti-oxidation. Material Basis of Spica Prunellae with chemoprevention and anti-lung cancer activity was studied from integrated and systematic points.This thesis was first to study the documents of the progress in drug screening of chemoprevention and anti-cancer and the Spica Prunellae chemical composition and pharmacological effects at home and abroad, and then carried out following the experimental study:(1)Firstly, Spica Prunellae material basis structural units were separated by grade, and the characterization of the structural units were represented useing of UV and high-performance liquid chromatography methods.We made an systematic analysis about the structural units of four extracts (by 95%, 60%, 30% ethanol and water gradient extract) of Spica Prunellae, and determined the content of total flavonoids, total phenols and total triterpene. The content of ursolic acid and oleanolic acid in 95% ethanol extract were determined.(2) Secondly, the structural units of Spica Prunellae on the prevention and treatment of lung cancer in vitro screening experements and its compatibility activities were studied.MTT assay was used to investigated the influence of different extracts from Spica Prunellae on antiproliferative effect of A549 and SPC-A-1 human lung cancer cells in vitro. 60% ethanol extract has the best antiproliferative effect with the lowest IC50 for single administration on two cells, 95% ethanol extract was followed. When 60% ethanol extract was used for compatible administration, the antiproliferative effect was bigger than its single administration. Four structural units for compatible administration on two cells has the best antiproliferative effect with the lowest IC50. The effect of three structural units for compatible administration were not better than that of 60% ethanol extract compatible administration with 95% ethanol extract. From the screening point, 60% and 95% ethanol extracts were effective structural units of Spica Prunellae on antiproliferation of human lung cancer cells.(3) Thirdly, the structural units of Spica Prunellae on the prevention and treatment of lung cancer in vivo screening experements were studied.The mouse modles bearing tumor were constructed by injecting subcutane- ously Lewis mouse tumor cells, then were treated with 95%ã€60%ã€30% and water extracts from Spica Prunellae to observe the weight of tumor to evaluate their anti-tumor activity in vivo.The results showed that 60% ethanol extract and 95% ethanol extract had good anti-tumor activity.(4) Lastly, the structural units of Spica Prunellae on the prevention and treatment of lung cancer in antioxidant effects activities were studied.ABTS, DPPH and FRAP methods were applied to study antioxidant of different extracts from Spica Prunellae in vitro. 60% ethanol extract has the strongest antioxidant according to the high content of total flavonoids and total phenols.By the screening exprements in vitro and in vivo, conbined with antioxidant in vitro, 60% ethanol extract and 95% ethanol extract were the effective Structural Units of Spica Prunellae for chemoprevention and anti-lung cancer activity. We supposed that such as triterpene, flavonoids and phenols may be the material basis of Spica Prunellae for chemoprevention and anti-lung cancer activity. |
