Rheumatoid arthritis(RA) is one of serious chronic diseases which threat Public healthy in the world, the mechanism of outbreak is complicated. The drug of total triterpenoids from Schefflera arboricola Hayata was selected as the research object; and then we evaluated the anti-rheumatoid arthritics effect as well as anti-rheumatoid arthritics mechanism of the drug. The study included five parts:1. The extraction and purification technology of total triterpenoids from Schefflera arboricola Hayata was investigated. With the content of total triterpenoid as index, the orthogonal test was applied to determine the optimal extraction condition. The optimum condition of extraction technology was 10 times of 95% ethanol extracted for 60 min, and extracted once. The optimal purification conditions were that: the concentration of sample at natural pH level was 0.884 mg/mL, the maximum loading amount was 6 BV as well as solvent of elution was 80% of alcohol(8BV).2. The anti-inflammatory activities of total triterpenoids from Schefflera arboricola Hayata was discovered by means of the utilization of the following models: Xylene-induced ear edema, CMC-Na induced leukocyte transfer in mice. The results indicated that the ear edema and leukocyte transfer can be highly restrained by total gatedtriterpenoids from Schefflera arboricola Hayata. It is clear that total triterpenoids from Schefflera arboricola Hayata has a remarkable anti-inflammatory activities.3. The analgesic effects of total triterpenoids from Schefflera arboricola Hayata was investigated by establishing the model of ethylic acid induced writhing response in mice. The experiments showed a clear inhibition of the pain reaction induced by ethylic acid in mice and a better analgesia than troche of Schefflera arboricola Hayata.4. The adjuvant-induced arthritis(AA) rats models were utilized to study the mechanism of resistance against RA of total triterpenoids from Schefflera arboricola Hayata through the following experiments: paw swelling in rats, index of RA, index of immune organs, the inflammatory cytokines in serum, the level of Cor and CIC. The experiments demonstrated that total triterpenoids from Schefflera arboricola Hayata have a strong effect on improving the situation of paw swelling and recover the immune function of AA rats. Restrictions the secretion of Cor and CIC in AA rats was also observed, indicating the anti-rheumatoid arthritis activities.5. With stimulating of LPS, the RAW264.7 macrophages were be the basis in this explore, we established models of inflammation and researched the anti-inflammatory activity and pharmacological mechanism of anti-inflammation of total triterpenoids from Schefflera arboricola Hayata and its active ingredients by ELISA and Griess in vitro.The results suggest that total triterpenoids from Schefflera arboricola Hayata can antagonize the damage of LPS induced inflammatory model in vitro.Total triterpenoids from Schefflera arboricola Hayata have great effects in the anti-inflammation and analgesic. It shows a great function in the anti-rheumatoid arthritis through the improvement of many Inflammation index of AA rats. The mechanism of the anti-inflammation effect is the balance control of the network of cytokines. |
