Objectives To established the rat model of post-traumatic stress disorder(PTSD), detect the protein and m RN A expression level of Beclin 1 and LC-3 in hippocampus and amygdala, and explore the mechanisms volume changes in hippocampus and amygdala of PTSD.Methods 80 adult male rats were randomly divided into control group(n=20) and model group(n=60). Single inescapable electric foot shock after SPS procedures(SPS & S) was used to establish PTSD rat model. Open- field test, elevated plus maze test and Morris water maze test were used to detect the rats’ behavioral change. Then rats were sacrificed at the subsequent 1, 4, 7, 14 and 28 d. The expression of Beclin1 and LC-3 m RNA and protein in hippocampus and the amygdala were detected using RT-PCR and Western-blotting respectively. Statistical analysis was performed us ing the SPSS 17.0 statistics software.Results 1, Open-field test results show that PTSD rats walking distance, time spent in central, and times upright were lower than control rats(P <0.05). The average escape latency of PTSD rats were higher than control rats(P <0.01). And percentage of time spent in the open arms of PTSD rats was significantly lower than control group(P<0.01). 2, Expression of Beclin1 and LC-3 protein increased in hippocampus and amygdala of PTSD rats(P<0.01), and 7d peaked(P <0.05). 3, Expression of LC-3 m RNA and Beclin1 m RNA in hippocampus increased in PTSD 1d, 7d peaked and 28 d still higher than control rats(P <0.05). In amygdala, compared to control rats, expression of Beclin 1 m RNA decreased in PTSD 1d and 28 d, while increased in PTSD 4d, 7d and 14d(P <0.05). The expression level of LC-3 m RNA lower than control rats in PTSD 1d, 4d, 14 d and 28 d, higher in PTSD 7d(P<0.05).Conclusions 1, SPS & S stress is a good animal model of PTSD; 2, Protein and m RNA expression of Beclin 1 and LC-3 in PTSD rat hippocampus and amygdala changed, which may be related to hippocampus and amygdala neuronal autophagy in PTSD rats.
