Objective: To explore the fast and efficient Chinese wolfberry effective components extraction method, this paper intends to pass on the basis of the extraction of biological enzyme hydrolysis, use ultrafiltration and purification technology, study the application of the integrated technology of biological enzyme hydrolysis extraction and ultrafiltration purification technology in the extraction of Lycium barbarum polysaccharides, in order to improve the utilization rate of Chinese wolfberry, expand Chinese wolfberry medicinal and health value.Methods: In order to improve the extraction rate and utilization rate of LBP total flavonoids, this paper combines the enzyme solution with the traditional extraction process and use ultrafiltration purification, through the uniform design optimize different kinds of enzyme hydrolysis extraction process conditions of LBP, and antioxidant activity as the evaluation index, the enzyme assisted extraction process and extraction liquid was studied,and on this basis, using compound enzyme to orthogonal experiment, with the compound enzyme assisted extraction of LBP and optimum technological conditions of total flavonoids; Star design- response surface method is used to select optimum parameters of LBP ultrafiltration purification. In order to establish can reflection of Chinese wolfberry "internal", "comprehensive" medlar enzyme solution assisted extraction method, To provide a certain theoretical basis for the industrialization of the production of wolfberry fruit rich in wolfberry liquid food and liquid medicine.Results: 1.The technological conditions for the extraction of the polysaccharide from the enzyme were as follows: the dosage of cellulase was 0.15%,Enzymatic hydrolysis temperature of 45 ℃, enzymolysis time 1.5h, ph4.4, the dosage of pectinase enzyme was0.15%, enzymolysis temperature of 45℃, enzymolysis time 1.5h, pH5.0; dosage of papain0.15%, enzymatic hydrolysis temperature of 55℃, enzymolysis time1.0h, pH4.6; dosage of xylanxylanase enzyme was 0.15%, enzymolysis temperature55 ℃, enzymolysis time1.5h,pH4.4; 2.compound enzyme assisted extraction technological conditions of Lycium barbarum polysaccharides: dosage of compound enzyme was 0.2%, hydrolysis temperature of 50℃,enzymatic hydrolysis time was 2H. 3. Enzyme assisted extraction technology of totalflavonoids from conditions:0.1% compound enzyme dosage, the best technological parameters of enzymatic hydrolysis time was 1.5h, enzymolysis temperature of 40℃ 4. The central composite design and response surface method optimization of Lycium barbarum polysaccharide ultrafiltration:Ultrafiltration temperature 30 degrees, ultrafiltration pressure0.11 Pa, solid-liquid ratio 1:15.Conclusion: biological enzyme solution extraction technology can significantly increase in medlar mainly effective group of mass transfer and improve the rate of polysaccharide and total flavonoids, by ultrafiltration purification technology to obtain the high purity of LBP;and for the industry of production are rich in Lycium barbarum polysaccharide wolfberry liquid food and Chinese wolfberry liquid agent provides a theoretical basis. |
