Experimental Research On STIP1’s Expression In Ovarian Tissues And Blood Serum Of Patients With Ovarian Tumors | Posted on:2016-05-26 | Degree:Master | Type:Thesis | Country:China | Candidate:Z B ( L u c k r e s s e A D | Full Text:PDF | GTID:2284330503977564 | Subject:Obstetrics and gynecology | Abstract/Summary: | PDF Full Text Request | [Objective]The aim of our study is to evaluate and compare the expression of stress-induced phosphoproteinl (Stipl) in the blood serum and ovarian tissues of patients with benign ovarian tumors from patients with ovarian cancer and healthy ovaries, and also understand the expression of Stipl in benign ovarian tumors patients.[Methods]Through a sandwich enzyme immunoassay (ELISA) Stipl serum plasma level was measured in fifteen patients with endometrial cyst; fifteen patients with others benign ovarian tumor; ten primary ovarian cancer patients (stage I-IV) and ten patients with uterine fibroid and healthy ovaries. Then STIP1 was immunohistochemically analyzed in the same fifty patients ovarian tissues after surgeries. This staining technique allow a visualization of tissues constituents localization under microscope through a two steps process including antigen-primary antibody interactions and the detection of bound antibody by a chromogen. Continuous variables are descriptive using mean+sd; One sample k-s test is applied to check data distribution, T-test were used for continuous variables, one-way ANOVA followed by a LSD test were used to compare the results of Elisa and IHC and for a comparison between the groups. The diagnostic accuracy of CA125 and stipl biomarker and cut-off points were assessed by building of the receiver operating characteristic ROC).Statistical analysis were performed using Statistical package for the social sciences (SPSS) version 20.0. For all analyses, p values less than 0.05 (p<0.05) were considered statistically significant.[Results]Stipl expression in cancer group is significantly higher compare to the other three groups(p<0.05).Stipl serum level is significantly highly express in the endometrial cyst group compare to stip1 serum level in benign tumor group (p=0.004). The mean value of Stip1 serum level in endometrial cyst group is 3.58 (n=15) while it is 2.02(n=15) in benign tumor group. Stipl is significantly express in endometrial tissue compare to benign tumor group(p=0.001).Additionally data analysis of patients as a total group showed that Stipl biomarker sensitivity(100%) with area under the curve(AUC= 0.818±0.081) is higher then CA125 biomarker sensitivity(93.3%) with area under the curve(AUC=0.902±0.054).In contrast,specificity of CA 125(73.3%) was higher than Stipl(53.3). The cut off value for Stip1 serum value is 7.10,meaning that when serum level of stipl in a patient with suspected adrenal masse is above 7.10, she has a relative higher risk of having endometrial cyst, but when the patient stipl serum level is less than 7.10 than the patient is more likely to be diagnosed with a benign ovarian tumor.[Conclusions]Stipl expression in cancer group is significantly higher compare to the other three groups. In both methods, IHC and ELISA, Stipl is highly express in patients in with endometrial cyst compare to patients with others benign ovarian tumors.
