The effects of ORI on the proliferation and apoptosis of 143 B cells were analyzed by crystal violet staining, flow cytometry and Western-blot.2. The effect of ORI on PI3K/Akt signal in 143 B cells was analyzed by Western blot.3. The effect of ORI on PTEN in 143 B cells were analyzed by Western blot.4. The effect of ORI on the proliferation of 143 B cells was analyzed by the crystal violet staining after silence or over expression of PTEN.5. Western blot was used to detect the influence of silence or over expression of PTEN on PI3K/Akt signal.6. The effect of PTEN and ORI on the ectopic osteosarcoma was detected in vivo.7. To detect the effect of PTEN on Wnt/ β-catenin signal in 143 B cells by blot Western.Results1. ORI could inhibit the proliferation and induce apoptosis of 143 B cells in a concentration dependent manner.2. ORI could inhibit the expression of PI3K/Akt signal in 143 B cells.3. The expression of PTEN in 143 B cells increased with the concentration of ORI.4. After silenced or over-expressed of PTEN, the proliferation of 143 B cells was stimulated or inhibited correspondingly.5. After silenced or over-expressed of PTEN, the PI3K/Akt signal in143 B cells were the increased or inhibited correspondingly.6. ORI and PTEN could inhibit the growth of the tumor, and inhibition of PTEN could promote tumor growing.7.
