| Objective:To explore the effect and molecular mechanism of mono-ADP-ribosylation of histone H3 Arginine on invasion of human colon LOVO cells.Methods: In this experiment, Un-transfection group and GFP-Vector group were set as control group, Arg117 Ala group and Arg117 Lys group were set as experimental group. Expression of H3K9me2,Runx3,β-catenin and MMP7 were tested by western blot. The m RNA leveles of Runx3 was detected by Quantitative real time polymerase chain reaction(QPCR). Ability of invasion was determined by Transwell invasion assay.Results:(1)Effect of mono-ADP-ribosylation of histone H3 Arginine on the invasion of LOVO cells : Ability of invasion was tested by Transwell invasion assay. The experimental results show that the number of cells which move through matrigel was decreased significantly in Arg117 Ala group and Arg117 Lys group( P<0.01). No statistical significant difference in Arg117 Ala group and Arg117 Lys group(P>0.05).(2)The molecular mechanism of mono-ADP-ribosylation of histone H3 Arginine in human colon LOVO cells invasion: The expression of H3K9me2,Runx3,β-catenin and MMP7 in each group were detected by western blot. There were no significant differences in Arg117 Ala group and Arg117 Lys group(P>0.05).The m RNA level of Runx3 was increased in Arg117 Ala group and Arg117 Lys group, compared with Un-transfection group and GFP-Vector group(p<0.01). There were no significant differences in Arg117 Ala group and Arg117 Lys group(P>0.05).Conclusion:This study reveals that mono-ADP-ribosylation of histone H3 Arginine has an effect on the invasion of LOVO cells. The concrete mechanism might be it up-regulate the expression of H3K9me2, and then downregulate the expression of Runx3 which increased the expression of MMP7. Hence, mono-ADP-ribosylation of histone H3 Arginine may play an important role in invasion of colon cancer. However, the detailed mechanism is still needed to further research. |
