Objective : It is creative to choose zebrafish as a whole animal model and investigating the inhibition effect of Fufang Kuh-seng Injection and its combination with cisplatin to angiogenesis. The experimental results will provide experimental evidence to the TCM and chemotherapy drugs used in combination to the treatment of osteosarcoma. Meanwhile, this study is also a useful attempt to explore the issue about chemotherapy cisplatin attenuated efficiency.Method and Results : Fufang Kuh-seng Injection set three dose(2.5, 6.25,12.5μL/m L) to treat AB zebrafish. The results show that in high dose group(12.5 mu L/mL), the number of functional internodes vascular that get blood through decreased significantly. Compared with the control group, the difference is significant(p < 0.05). To dye the zebrafish blood vessels using alkaline phosphatase staining, it is found that the Fufang Kuh-seng Injection has obvious influence on functional blood vessels of zebrafish(graph 3). 2、At low doses(25 to 50 mu g/mL of Fufang Kuh-seng Injection, matrine had no obvious effect on zebrafish angiogenesis.When dose increases to 100 mu g/mL, it shows obvious inhibitory effect(p < 0.05).there is no obvious influence of oxymatrine within the experimental dose scope. 3、In50 mu g/mL doses, Cisplatin donot significantly affect the zebrafish internode angiogenesis. After adding Fufang Kuh-seng Injection(5 mu L/mL), the two produce obvious inhibition to the ISV of zebrafish, compared with the control group, it has significant difference(p < 0.05); with the increasing of dose of Fufang Kuh-seng Injection, the synergy enhances(p < 0.01).Conclusion : In this experiment, compared with single use of the two, the combination of Fufang Kuh-seng Injection and cisplatin plays a more obvious inhibitory effect in zebrafish internode angiogenesis. It shows that the combination has synergy effect.Most of the chemotherapy drugs have different degrees of bone marrow suppression. Reducethe use of chemotherapy drug dose is valuable for cancer therapy.Because inhibition of the combination of Fufang Kuh-seng Injection and cisplatin totumor angiogenesis enhanced, so the cisplatin can be used in low doses, the benefits is that the side effects of cisplatin become less and tolerance of patients enhanced,that can make chemotherapy smoothly, also indirectly increase the curative effect.This study also provides a new method of traditional Chinese medicine combined with chemotherapy. In the process of the research of Chinese traditional medicine, it often faced with difficult problem that the complex composition system is difficult to explain, pharmacological effects can’t be comprehensive characterized and so on.With the combination, the process is more complex. As a new model, Zebrafish can simulate some of the characteristics of physiology and pathology of life.(such as angiogenesis, etc.)On the overall level, compared with the commonly used mammals,the combination greatly simplifies the experiment link and cost and could be an ideal alternative.
