| Objectives: This paper aims to observe the effects of Shengui Baoxin prescriptions on the contents of Left ventricular mass index(LVMI), N-Terminal-pro-B-Type Natriuretic Peptide(NT-pro BNP), Succinate dehydrogenase(SDH), Na+-K+-ATPase,Ca2+-Mg2+-ATPase in rats with heart failure; to explore the effect of the drug on the energy metabolism of myocardial cells in rats.Methods: 50 clean grade male Wistar rats were selected from Specific pathogen Free(SPF), and 10 randomly divided into blank control group with routine feeding throughout the entire process. The 40 rats were taken the method of subcutaneous multi-point injection of isoprenaline(ISO), and each rat was given is ISO(85mg/kg·d). After continuous modeling for 2 days, the rats were randomly divided into 4 groups: model group, Shengui Baoxin prescriptions high dose group, middle dose group, low dose group. Subsequently, model groups were given the same dose of saline, Shengui Baoxin prescriptions group was given corresponding concentration of traditional Chinese medicine orally according to high, medium and low dose. The rats were executed after 8 weeks′, and calculate the group of 5 rats LVMI. Immuno fluorescence method was used, to determinate the content of NT-proBNP in rats, and the UV spectrophotometer detected the myocardium SDH, Na+-K+-ATPase and Ca2+-Mg2+-ATPase activity.Results:(1) LVMI: compared with the blank group, the rest of the group increased(P<0.01). Compared with the model group, high dose group has difference(P<0.01). Compared with the low dose group, high dose group has difference(P<0.01).(2) The level of NT-pro BNP in rats: compared with the blank group, the rest of the group increased(P<0.01). Compared with the model group, high dose group and middle dose group and low dose group had difference(P<0.01). Compared with the low dose group, the high dose group were different(P<0.01).(3) SDH: compared with the blank group, the rest of the group reduced(P<0.05). Compared with the model group, high dose group and middle dose group and low dose group had difference(P<0.05). Compared with the low dose group, the high dose group were different(P<0.05).(4) Na+-K+-ATPase: compared with the blank group, the rest of the group reduced(P<0.01). Compared with the model group, high dose group and middle dose group had difference(P<0.05). Compared with the low dose group, the high dose group were different(P<0.01).(5) Ca2+-Mg2+-ATPase: compared with the blank group, the rest of the group reduced(P<0.05).Compared with the model group, high dose group and middle dose group and the low group had difference(P<0.05). Compared with the low dose group,the high dose group were different(P<0.05).Conclusion: Shengui Baoxin prescriptions can inhibit myocardial remodeling in rats with chronic heart failure and improve cardiac function in heart failure rats. The treatment of heart failure pathway may be by increasing the enzyme activity during myocardial energy metabolism and the adenosine triphosphate(ATP) production, leading to the improvement of myocardial energy metabolism disorder. The concentration of the drug is mainly in the concentration of high dose. |
