Corydalis Pallida is the whole plant of poppy plant tooth buds yellow violet, and Tibetan Codonopsis Nervosa is the whole plant or the aboveground part of the genus Codonopsis Pilosula. Corydalis Pallida and Codonopsis Nervosa are the main ingredients of Tibetan medicine Luodui Dorje Particles, and Tibetan medicine Luodui Dorje Particles is mainly using for the treatment of gout. Since we do not have the report about the effect of activity of Corydalis Pallida, and Codonopsis Nervosa towards to activity of Xanthine Oxidase and the main chemical compositions of Corydalis Pallida, I have chosen Corydalis Pallida and Codonopsis Nervosa as my research objects. I will measure the activity of Xanthine Oxidase and identify the main chemical compositions of Corydalis Pallida so that I can do some materially basic research for the study of Tibetan medicine about the treatment of gout. Specifically including the following aspects: 1. The effect of Tibetan Codonopsis Nervosa towards the Activity ofXanthine OxidaseObjective: to study the effect of the extract and monomer of Tibetan Codonopsis Nervosa towards the activity of Xanthine Oxidase. Method: make, steam and dry methanol extracting solution of Tibetan Codonopsis Nervosa. Dilute the extract and monomer of Tibetan Codonopsis Nervosa into different concentrations with buffer solution, and then I will use ultraviolet spectrophotometry to measure the effect towards the activity of Xanthine Oxidase. Result: both the methanol extracting solution and the purified monomer of Tibetan Codonopsis Nervosa have inhibiting effect towards Xanthine Oxidase, and it shows us a good concentration-response relationship. 2. The effect of Luodui Dorje Particles and its Main CompositionsCorydalis Pallida towards the Activity of Xanthine Oxidase.Objective: to research the effect of Luodui Dorje Particles and its chemical Compositions towards the activity of Xanthine Oxidase. Method: make respectively methanol extract of Luodui Dorje Particles and Corydalis Pallida’s methanol, acetone and water extract; dilute into different concentrations with buffer solution, and compare with common sage herb in positive control, then use ultraviolet spectrophotometry to measure the effect of all kinds of extracts towards the activity of Xanthine Oxidase. Result: Luodui Dorje Particles and all the extracts of Corydalis Pallida have obviously inhibiting effect towards Xanthine Oxidase 3. The Isolation and Identification of Chemical Compositions ofCorydalis PallidaObjective: to study the chemical compositions of Corydalis Pallida. Method: weigh dry herbs 20 kg, and then get the dry extract through reflux extraction. Extract the dry extract with different solvents to get different parts of extracts. Get different compounds through using thin layer chromatography and various means of silica gel column chromatography to separate the different parts of extracts. Then I will analyze and identify the compound’s structure according to physicochemical property and spectrum analysis. Result: separate and extract thirteen elements from the parts of ethyl acetate and dichloromethane of Corydalis Pallida.
